| Objective To investigate the metastasis mechanism of lung cancer with spleen deficiency,and the mechanism of Jiawei Sijunzi in lung cancer with spleen deficiency.Method Given by multifactor method combined with cold and bitter purgation,fatigue and restricting food intake,the model of Lewis lung cancer mice with spleen deficiency was successfully established.Each group was dealt with the according plans and generally observed at the same time.On the 17th day,the blood taken from mice’s eyeballs was gauged for the serum gastrin;the lung tissue was taken for gauging IL-6 in the lung tissue homogenate.After the model was testified,mice were administrated with Jiawei Sijunzi for 14 days and then were sacrificed after weighed,being taken for lung,thymus and tumor tissues.Counting lung metastatic nodules,weighing the thymus and calculating the thymus index,gauging the gene expression of Fas and Bcl-2 by Real-time PCR.Result The model of Lewis lung cancer mice with spleen deficiency was successfully established.The general condition of mice with spleen deficiency was bad,and the serum gastrin were significantly lower and the IL-6 in the lung tissue homogenate were significantly higher than the normal group.The thymus index of lung cancer group and Jiawei Sijunzi group were higher than lung cancer with spleen deficiency group,but the pulmonary metastatic nodules were less.The expression of Fas mRNA of lung cancer with spleen deficiency group were lower than lung cancer group and Jiawei Sijunzi group.The expression of Bcl-2 mRNA of lung cancer with spleen deficiency group were higher than lung cancer group and Jiawei Sijunzi group.Conclusion Spleen deficiency decrease the thymus index,increase the expression of IL-6,provide an appropriate inflammatory microenvironment for the growth and metastasis of lung cancer.Spleen deficiency downregulate the expression of Fas mRNA,upregulate the expression of Bcl-2 mRNA,promote the metastasis of lung cancer.Jiawei Sijunzi reverse the spleen deficiency,protect the immune organs(thymus),improve immunity,upregulate the expression of Fas mRNA,downregulate the expression of Bcl-2 mRNA,inhibit lung cancer metastasis. |
