| Objective : To observe the changes of liver related cytokines in process of solution fiber Decoction in the treatment of liver fibrosis CTGF,TGF-beta 1,PDGF and CTGF m RNA、TGF-β1 m RNA 、 PDGF m RNA.Study of Huganjiexian Decoction in the treatment of liver fibrosis and the mechanism.Methods : Rat models of hepatic fibrosis were made by peritoneal injection of carbontetrachloride for 12 weeks.Model control group underwent the modeling process.Xiaochaihutang positive drug control group from the nine weeks,pay Xiaochaihutang treatment.Huganjiexian Decoction of high,middle,low concentration were given to rats of different groups respectively from the ninth week.Normal control,model control,Minor Decoction of Bupleurum groups were made as controls.Immunohistochemical staining was executed for detecting expression of CTGF,TGF-β1,PDGF in liver tissues.CTGF m RNA 、TGF-β1 m RNA、PDGF m RNA were detected by in situ hybridization.Professional software for image analysis was used.Results :1.1 In Normal control group and model control group,liver tissue CTGF expression were 0.0024±0.0025、0.0511±0.0244,low doses of Huganjiexian Decoction group,middle dose of Huganjiexian Decoction group,high dose of Huganjiexian Decoction group,Xiaochaihutang group expression were 0.0288±0.0283、0.0221±0.0125、0.0077±0.0060、0.0045±0.0022.1.2 In Normal control group and model control group,liver tissue TGF-β1 expression were0.0035±0.0029、0.0491±0.0218,low doses of Huganjiexian Decoction group,middle dose of Huganjiexian Decoction group,high dose of Huganjiexian Decoction group,Xiaochaihutang group expression were 0.0169±0.0109、0.0124±0.0078、0.0097±0.0070、0.0069±0.0050.1.3 In Normal control group and model control group,liver tissue PDGF expression were0.0036±0.0025、0.0231±0.0108,low doses of Huganjiexian Decoction group,middle dose of Huganjiexian Decoction group,high dose of Huganjiexian Decoction group,Xiaochaihutang group expression were 0.0197±0.0121、0.0080±0.0048、0.0066±0.0038、0.0060±0.0046.2.1 The expression of liver tissue CTGF m RNA in the normal control group,model control group,low doses of Huganjiexian Decoction group,middle dose of Huganjiexian Decoctiongroup,high dose of Huganjiexian Decoction group,Xiaochaihutang group were 0.0132±0.0078、0.0269±0.0150、0.0177±0.0068、0.0150±0.0097、0.0098±0.0061、0.0147±0.0038.2.2 The expression of liver tissue TGF-β1 m RNA in the normal control group,model control group,low doses of Huganjiexian Decoction group,middle dose of Huganjiexian Decoction group,high dose of Huganjiexian Decoction group,Xiaochaihutang group were 0.0269±0.0145、0.0906±0.0406、0.0812±0.0353、0.0393±0.0232、0.0278±0.0180、0.0193±0.0087.2.3 The expression of liver tissue PDGF m RNA in the normal control group,model control group,low doses of Huganjiexian Decoction group,middle dose of Huganjiexian Decoction group,high dose of Huganjiexian Decoction group,Xiaochaihutang group were 0.0291±0.0133、0.0687±0.0278、0.0648±0.0244、0.0344±0.0211、0.0402±0.0204、0.0245±0.0129.Compared with model group,expressions of CTGF,TGF-β1,PDGF and m RNA in liver tissue of rate of high dose of Huganjiexian Decoction decreased significantily(P < 0.01);expressions of TGF-β1 in liver tissue of rate of middle dose of Huganjiexian Decoction decreased(P < 0.05),PDGF decreased significantily(P < 0.01),but CTGF has no statistical difference significance(P> 0.05);expressions of CTGFm RNA in liver tissue of rate of middle dose of Huganjiexian Decoction decreased significantily(P < 0.05),TGF-β1m RNA and PDGFm RNA decreased significantily(P < 0.01);expressions of CTGF、TGF-β1、PDGF and m RNA in liver tissue of rate of low dose of Huganjiexian Decoction has no statistical difference significance(P> 0.05).Conclusion: Huganjiexian Decoction could reduce expressions of promoting fibrosis Cytokines CTGF,TGF-β1,PDGF and CTGF m RNA、TGF-β1 m RNA、PDGF m RNA.This maybe the important mechanisms of the rapeutic effects of Huganjiexian Decoction of fibrosis. |
