| Objective:To investigate the chemical constituents of the aerial parts of Clerodendranthus spicatus,which was extracted by 80%ethanol,isolated,purified and structure identified.Methods:To isolated these compounds by means of different chromatographic methods such as RP-18,Sephadex LH-20,MCI gel CHP-20P and HPLC.The structures of these compounds have been confirmed by spectroscopic methods(1H-NUR,13C-NMR,MS,2D-NMR,UV,CD,X-Ray)and VCD calculation.Results:47 compounds were isolated from Clerodendranthus spicatus,they were identified as(4E,9S)-9-Hydroxy-2,4-megastigmadien-1-one(1),3-Hydroxybutyl-2,4,4-trimethyl cyclohexa-2,5-dienone(2),Vomifoliol(3),Loliolide(4),6-Hydroxy-1-oxo-7,8-dihydro-β-ionone(5),Alkaloid 1(6*),Diterpenoid 1(7),Diterpenoid 2(8),Orthosiphol W(9),Neoorthosiphol A(10),Orthosiphols R(11),Orthosiphols L(12),Orthosiphols A(13),Orthosiphols K(14),Orthosiphol B(15),Orthosiphol O(16),Orthosiphol A(17),Neoorthosiphol B(18),Orthosiphol D(19),2-O-Deacetylorthosiphol J(20),Orthosiphol M(21),Orthosiphol N(22),Orthosiphonone A(23),Orthosiphol E(24),Norstaminol S(25),Spicatusoid A(26*),Vitexnegheterion H(27),Spicatusoid B(28*),Spicatusoid C(29*),Spicatusoid D(30*),Spicatusoid E(31*),Euscaphic acid(32),2α,3β,19β-Trihydroxyurs-12-en-28-oic acid(33),Arjunolic acid(34),Lignin 1(35*),(±)-Rosmarinsauremethylester(36),trans-7,8-Dihydro-(4’-hydroxy-3’ 5’-dimethoxyphenyl)-1,3-dimethoxy-6,7-dimethyl-2-naphthol(3 7),Vinyl acetate(38),Kaempferol 3-O-α-L-rhamnopyranoside(39),5,7,8,4’-Tetramethoxyflavanone(40),5-Hydroxy-7,3’4’-trimethoxyflavone(41),5,7,8,3’,4’-Pentamethoxyflavanone(42),Ethyl coumarate(43),3’,4’,5,6,7-Pentamethoxyflavone(44),4’-Methoxy-5,6,7-trimethylbaicalein(45),Fragransin B1(46),Fragransin B2(47).Conclusion:The types of 47 compounds have been identified relate to sesquiterpenes,diterpenes,triterpenes,phenolic acids.Compounds 6、26、28-31、35 were new compounds,compounds 2 was a new natural product.Above these compounds,not only deepened the chemical understanding of this herbal medicine,but provided a new chemical material foundation for further rational development and utilization.At the same time,cytotoxic assay(HL-60,SMMC-7721,A-549,MCF-7 and SW-480),cell viability assay(NRK-49F),TGF-β1-stimulated proliferation inhibitory assay(NRK-49F)were used to evaluate their biological activities,some of them present positive reactions towards these assays. |
