Objective To approach the protecting effects of aqueous extract from a herb C.Spicatus on chronic renal failure rats induced by adenine, as well as the possible mechanism involved in.Methods The animal model of CRF was established by the adenine gastric infusion method with 200~220g Sprague-Dawley rats. The experimental rats were randomly divided into 5 groups. Group 1 was normal group. Group 2 was model control group. Group 3 was given the C.Spicatus decoction at the dosage of 2.5g·kg-1. Group 4 was given the C.Spicatus decoction at the dosage of 5.0g·kg-1 and Group 5 was Niaoduqing positive group. After therapying seventeen days,detecting the contents of urine uric acid.Continuing and therapying 4 weeks, BUN,Cr,MDA of blood serum were respectively detected, as well as determined GFR and renal plasma flow (deployed Inulin and PAH clearance rate formula calculation).The kidneys were observed for the histopathology by HE dyeing and detected the level of bFGF by immuhistochemistry dyeing.Results 1. Body weight of model control group and C.Spicatus groups were significantly lighter than those of normal control group (P<0.05) .Body weight of large dosage of C.Spicatus group(337.89±28.87g) was significantly higher than those of model control group(304.86±31.37g) (P<0.05)among the total. 2. The contents of urine uric acid of C.Spicatus groups and Niaoduqing positive group were significantly higher than those of model control group(7.44±1.87μmol·L-1), (12.19±1.87μmol·L-1 P<0.05) in large dosage of C.Spicatus group, (8.47±1.24μmol·L-1 P<0.05) in small dosage of C.Spicatus group and (8.23±1.41μmol·L-1 P<0.05) in Niaoduqing positive group among the total,but there were no significant in each therapying group. 3. The BUN of blood serum of C.Spicatus group and Niaoduqing positive group were lower than those of model control group (26.89±5.86mmol·L-1) , (17.72±3.44 mmol·L-1 P < 0.05) in large dosage of C.Spicatus group , (15.95±3.46 mmol·L-1 P < 0.05) in small dosage of C.Spicatus group and(18.05±4.96mmol·L-1 P < 0.05) in Niaoduqing positive group, but there were no significant in each therapying group. 4. The inulin and PAH clearance rate of C.Spicatus groups and model control group were significantly lower than those of normal control group(P<0.05),while the inulin clearance rate of large C.Spicatus group(0.70±0.08ml·min-1·kg-1) was significantly higher than those of model control group(0.47±0.08 ml·min-1·kg-1)(P<0.05). 5. The level of MDA of C.Spicatus groups and Niaoduqing positive group were lower than those of model control group(7.86±0.71nmol·mL-1) , (6.56±0.87nmol·mL-1 P <0.05) in small dosage of C.Spicatus group , (6.40±0.53nmol·mL-1 P <0.05) in large dosage of C.Spicatus group and (6.32±0.65nmol·mL-1 P <0.05) in Niaoduqing positive group. 6. The level of kidney weight/BMI of large C.Spicatus group was lower than those of model control group(3.53±0.30w-1·10-3) , (2.46±0.46w-1·10-3 P < 0.05) in large dosage of C.Spicatus group. 7. The expression of bFGF in the renal tubule and interstitial cell of model control group was significantly increased (P < 0.01);while the expression of bFGF in the renal tubule and interstitial cell of large dosage of C.Spicatus group was significantly less (P < 0.05).Conclusion The aqueous extract from a herb C.Spicatus decoction at the range of dosage from 2.5g·kg-1 to 5.0g·kg-1 has effect on the kidney of CRF rats induced by adenine. The effective mechanism may be associated with the its accrescence of uric acid evacuation,improvement of renal function and anti-oxydation activity. In addition, it might relate to inhibitory effect on the proliferation of fibroblast cells.
