Cerebral infarction,also known as ischemic stroke,is one of the common and frequently occurring neurological diseases.The middle cerebral artery is one of the main blood vessels in the brain,and the focal cerebral ischemia model can be obtained by occlusion of the middle cerebral artery.In this study,a mouse model of middle cerebral artery occlusion was successfully established by using a mouse model of severe immunodeficiency mice with both age and gender.The research laid the foundation for further study on the pathological changes of cerebral ischemia and drug efficacy.Methods:30 male SCID mice were randomly divided into sham operation group and MCAO operation group(n=15).The MCAO model was made by electrocoagulation and cut off the left middle cerebral artery in the SCID mice,making the MCAO model;the sham operation group underwent the same surgical procedure,but not do with the artery.Use laser speckle flowmetry real-time monitoring of cerebral blood flow in mice,and calculate cerebral blood flow ratio.The ratio of cerebral blood flow was calculated.The mice we re sacrificed neurological function after 24 hours of anesthesia.After 2 weeks,2,3,three 5-chloride phenyltetrazole(TTC)and diaminobenzidine(DAB)staining were used to calculate the percentage of cerebral infarction area.Results:1.The MCAO surgery group showed obvious neurological dysfunction,and the neurological function score was in accord with the actual performance.The sham operation group of rats without any symptoms.2.Compared with preoperative,the ratio of cerebral blood flow in MCAO group decreased significantly,and the ratio of cerebral blood flow was stable at 60 min after operation.3.Brain slices TTC,DAB staining showed significant cerebral infarction in the brain of mice in the MCAO group P sham operation group mice no such change.Conclusions:1.The model of cerebral ischemia model established in this experiment is consistent with the performance of MCAO model in behavior and pathology.2.This model can simulate the human cerebral infarction to a certain extent,the success rate is high,the infarct site is constant.The model is stable and reliable,and it is an ideal animal model for studying cerebral ischemic injury.
