Great progress has been made in the treatment of diabetes in recent years and the drugs used are mainly western medicine.These western medicine have significant hypoglycemic effect,clear targeted and hypoglycemic mechanism,while they bring up lots of side effects simultaneously.So relative researcher focused on new drugs of plant polysaccharide,which could have exact hypoglycemic effect and minimal side effect.Firstly,were constructed by means of injecting different concentration of streptozotocin with common feeding rats and low dose STZ with high fat and sugar feeding rats.And then we the weight,water and food intake,blood sugar,Insulin and molding rate were detected.Secondly,we extracted the Cynomorium Songaricum polysaccharide from Cynomorium Songaricum by using the method of hot water extraction.Finally,we established type 2 diabetic rat models using optimal modeling method.At the same time the experimental type 2 diabetic rats were randomly divided into four groups,positive control group,negative control group,Cynomorium Songaricum polysaccharide(200mg/kg)treatment group,glibenclamide(100 mg/kg)treatment group by intragastric injection with,Cynomorium Songaricum polysaccharide and glibenclamide,respectively,25 d and established negative control group with saline intragastric injection.Thus we investigated the effect of Cynomorium Songaricum polysaccharide to the the weight,water and food intake,blood sugar,blood fat,Total cholesterol estero(TC),Triglycerides(TG),High density lipoprotein(HDL),Low density lipoprotein(LDL),Insulin,Insulin receptor(INSR)and Insulin receptor substrate-1(IRS-1)of type 2 diabetic rats and explored the hypoglycemic effect and the mechanism of Cynomorium Songaricum polysaccharide in type 2 diabetic rats.The results are shown as follows:1)The results from the rats with common feeding showed that the weight in STZ(30,20mg/kg)group reduced significant after 15 days.The water intake in STZ(30,20 mg/kg)group increased after 10 days.the food intake in STZ(30,20 mg/kg)group increased after 15 days.The concentration of blood sugar from the three STZ injection group were increased after 10 days.The levels of insulin in STZ(30,20mg/kg)group decreased significantly.The molding rate from STZ(20,15 mg/kg),STZ(20,25 mg/kg)and STZ(30,20 mg/kg)groups were 10%,40% and 30%individually,the fourteen dead rats were all from STZ(30,20 mg/kg)group only.2)The results from the rats with high fat and sugar feeding showed that the weight in STZ(20,15 mg/kg)group reduced significant after 20 days,and the weight in STZ(30,20 mg/kg)group reduced significant after 10 days.The water and food intake,the concentration of blood sugar from the three STZ injection group were increased after 5 days.The levels of insulin in STZ(30,20 mg/kg)group decreased significantly.The molding rate from STZ(20,15 mg/kg),STZ(20,25 mg/kg)and STZ(30,20 mg/kg)groups were 30%,90% and 50% individually,the tenth dead rats were all from STZ(30,20 mg/kg)group only.3)The Cynomorium Songaricum polysaccharide significantly increased the weight of 2 type diabetes mellitus rats(P<0.01),and the effect was similar to the effect of glibenclamide(a hypoglycemic drug).The Cynomorium Songaricum polysaccharide significantly improved polydipsia,polyphagia,polyuria,which are caused by 2 type diabetes mellitus,and and the effect of polyuria was prior to the glibenclamide(P<0.01).The Cynomorium Songaricum polysaccharide significantly decreased the content of TC,TG,HDL in serum of 2 type diabetes mellitus rats(P<0.05,P<0.01),and significantly increased the content of LDL,and the effect was similar to the effect of glibenclamide(P<0.05).The Cynomorium Songaricum polysaccharide improved insulin levels of 2 type diabetes mellitus rats(P<0.05),the effect was similar to the effect of glibenclamide.The Cynomorium Songaricum polysaccharide significantly enhanced expression of INSR,IRS-1 protein(P<0.01),and compared with the effect of G complications of diabetes.In conclusion,compared with other induction methods,the method of high fat and sugar feeding with STZ(20,25 mg/kg)induce 2 type diabetic rat model has high molding rate and no death and can be used.Cynomorium Songaricum polysaccharide showed a promising hypoglycemic effect,improved polydipsia,polyphagia,polyuria,weight loss,lipid metabolism disorder and expression of INSR,IRS-1 protein significantly,which are caused by 2 type diabetes mellitus rats.The primary hypoglycemic mechanism may be related to following approach 1)to relieve the oxidative lesion of islet,protect the islet cell from injuring,promote the injured pancreatic islet to restore and enhance the exudation of insulin,hypoglycemic effect.2)to adjust lipid metabolism disorder,INSR,IRS-1 protein expression,protection of2 type diabetic rats diabetic organs to play its role in the overall protection of 2 type diabetic rats.Our study provided a strong theoretical support that the Cynomorium Songaricum polysaccharide could act as antihyperglycemic food and medicine against the complications of 2 type diabetes.
