Objective:To determine the effects of hydrogen sulfide on liver tissues in rats with type 2 diabetes.Methods:To buy 80 SD rats from Tongji medical college of HuaZhong science and technique university and the T2DM rat model was established by using high-sugar,high-fat diet and intraperitoneal injection of low-dose streptozotocin?STZ?.The model ratswererandomlydividedinto4groups?n=15each?;diabetic groups,diabetes+NaSH?anH2Sdonor?group,diabetes+DL-propargylgl-ycine[PAG,an inhibitor of cystathionine-?-lyse?CSE?]group,and normal control group.Rats were intraperitoneally injected with NaSH[56?mol/?kg.d?]and PAG[50mg?kg.d?]in DM+NaSH group and DM+PAG group,respectively,and with the same volume of normal saline in the DM and normal control groups.After 2 weeks,the serum levels of fasting blood glucose?FBG?and insulin,hydrogen sulfide concentrations in liver tissues were measured.The pathological changes of liver were observed under light microscopy staind with HE.The protein expression levels of insulin receptor substrate-2 in hepatocytes were detected by immunohistochemistry.The protein and mRNA expression levels of CSE?InsR and IRS-2 were determined by western blot and reverse transcription polymerase chain reaction?RT-PCR?.Results:The levels of FBG and plasma insulin are significantly reduced,the expression of CSE were incresed significantly and the expression levels of InsR and IRS-2 were decreased in liver tissue in DM group compared with normal control group.The levels of FBG and plasma insulin were also reduced and the expression levels of CSE?InsR?IRS-2 were slightly decreased in DM+NaHS group compared with DM group.Compared with DM group,the DM+PAG group had a increase in FBG?plasma insulin level and the expression level of InsR?IRS-2 and a huge decrease in expression level of CSE.Conclusion:H2S can promote insulin resistance through down-regulating expression level of InsR and IRS-2 in liver tissue of rats with type 2 diabetes and PAG can improve insulin resistance through up-regulating expression level of InsR and IRS-2 in liver tissue of rats with type 2 diabetes.
