ObjectiveTo study the effects of Liuweidihuang Decoction on Fasting blood glucose, Fasting serum insulin and Insulin sensitivity index of type2diabetic rats. Observated the morphological changes of Liver and Kidney, and the effects to secretions of Plantar Sweat Gland of all rats. Based on the above, we are continue to study the effects of Liuweidihuang Decoction on the expression of phosphorylated Insulin receptor substrate-2protein with the liver of the model rats. To simply discussed the mechanism of Liuweidihuang Decoction on type2diabetic rats with insulin resistance.MethodsEstablished the type2diabetic model with insulin resistance by feeding high sugar and fat diet, then intraperitoneal injection streptozotocin. Fasting blood glucose determined by Blood glucose meter, the level of Fasting serum insulin detected by Enzyme-linked immune-sorbent assay, then calculate Insulin sensitivity index according to ISI=In[1/FINS*FBG]. Hematoxylin-eosinstaining investigate the morphological changes of Liver and Kidney of all rats, and the changes of secretions of Plantar Sweat Gland. Immunohistochemistry detected the expression of phosphorylated Insulin receptor substrate-2protein with the liver of all rats.ResultsHigh sugar and fat diet feeding for four weeks conbined with intraperitoneal injection streptozotocin(45mg/kg) can result a rat model of type2diabetic model with insulin resistance, fasting blood glucose of the models were≥16.7mmol/L, insulin sensitivity index were declined(P <0.01). Liuweidihuang Decoction can declined Fasting blood glucose of type2diabetic rats with insulin resistance(P<0.01), and improve Insulin sensitivity index (P<0.01). The impaired Liver and Kidney of the model can be repaird after treatment, and its abnormal secretions of Plantar Sweat Gland had improved. Liuweidihuang Decoction could improve the expression of phos-phorylated Insulin receptor substrate-2protein with the liver of type2diabetic rats with insulin resistance(P<0.05).ConclusionThe impaired Liver and Kidney and the abnormal secretions of Plantar Sweat Gland can be improved after treatment by Liuweidihuang Decoction. Its mechanism may be related with the expression of phosphorylated Insulin receptor substrate-2protein improved after treatment. the expression of phosphorylated Insulin receptor substrate-2protein improved, Thus the IRS2protein of the Insulin signaling pathway had adjusted, And then improved the insulin signal transduction, and improved the IR, Finally improved the type2diabetic rats with insulin resistance.
