Objective: To investigate the effects of CAL-27 cell under the same dose of different ionizing radiation sources and identify the differently expressed microRNA(miRNA)for further study of radiotherapy in human tongue squamous carcinoma.Methods: Applied X-ray instrument and heavy ion accelerator irradiated CAL-27 cells;Flow cytometry was used for detecting the cell survival rate and apoptosis rate;High throughput sequencing technique(Hiseq)was applied for detecting the differentially expressed genes(DEGs).Via bioinformatic analysis,the main signal pathways which DEGs take part in were obtained.Results:1)The CAL-27 cell clone survival rate of Control,0.8 Gy X-ray group and 0.8 Gy heavy ion irradiation group were respectively(100±0.031)%?(93±0.031)%?(64±0.027)%(P<0.05),the apoptosis rate were respectively 1.9%,13.9%,and 30.4 %.2)Compared with X-ray irradiation,there were 33 miRNAs expression profile were changed after heavy ion irradiated,among them there were 12 miRNAs up-regulated(for example,hsa-miR-629-5p,hsa-miR-146b-5p and hsa-miR-15b-5p,et al.),21 miRNAs down-regulated(for example,hsa-miR-181a-2-3p,hsa-miR-130a-3p and hsa-miR-181a-3p,et al.)in CAL-27 cells by Hiseq and 4 novel miRNAs expressed differently(Novel-mir-519,Novel-mir-797,Novel-mir-577 and Novel-mir-624).3).15 pathways which DEGs participated in were screened out by KEGG database.Conclusions: Under the same dose condition,the heavy ion irradiation played a key role on biological effect of CAL-27 cells,and its mechanism of differently expressed miRNA may be associated with MAPK signaling pathways,which will lay a foundation for further study of miRNA in the molecular biology mechanism of tongue squamous carcinoma radiotherapy.
