Objective: Activity oriented,in view of the activity of a better one individual plant cultivation and identification of endophytic fungi,its fermentation product extraction and separation,in order to get the new structure and active monomer compounds.Structure of the monomer compounds identified,presumably,some students way of potential compounds,so that through the way of artificial cultivation or synthetic directional obtain large quantities of target product.The target product is likely to be with their host plants in xinjiang wei the same or similar active substances.Method: choose active strong strain identification,study its growth characteristics,different culture medium,temperature,illumination and pH 4 single factor’s influence on bacteria growth,optimized through the response surface test,bacterial species to determine optimal growth conditions.Then carries on the mass of the strain,fermentation by means of a series of chemical separation,the extraction and separation of the metabolites and ingredients to identification.Results:According to molecular sequencing and electron micrograph observe its morphology and spore characteristics,identification of the endophytic fungi alternaria.Under the condition of the dark,using potato culture medium(PDA),pH value of 7,temperature of 25 ℃environment,the species can be normal growth and reproduction,strain the average growth rate of 10.25 mm,d-1.In bulk after fermentation conditions for its metabolic product separation,21 compounds were separated and identified respectively 10 beta sitosterol,(2)the ergot,6,8 steroid-4(14),22 e-four ene-3 ketone;(3)phthalate butyl ester;(4)oleic acid;(5)methyl benzene formaldehyde;(6)linoleic acid;(7)2-oxabicyclo[3,2,2]nona-5,7,8-triene;(8)2-(4-hydroxyphenyl)ethyl acetate;(9)alternariol;(10)alternariol 4-methyl ether.Other compounds is still in identification.Conclusion: Based on the separation of the endophytic fungi of ferulic metabolites in xinjiang,found that for some activity is better and more valuable compounds,we can from molecular biology to study,and on the basis of its unique structural characteristics,further speculated that its biosynthetic pathway;From the aspects of gene,to a series of effective reorganization and knockout,cultivate engineering bacteria,in order to get the large quantities of target product,make these valuable compounds from quality to quantity of amplification,can fundamentally solve the separation of monomer compounds needed to sample weight,such as impurities,process more complicated problem,but also can improve the condition of shortage of monomer compounds in pharmaceutical research.
