ObjectiveTo observe the effects of sympathetic nerve transection on the level of IL(interleukin)-1?,IL-R?,IL-6,TNF(tumor necrosis factor)-?,NF(nuclear factor)-?B,TLR(toll-like receptor)4 in serum and spinal cord tissue of rabbits,which are pretreated by electroacupuncture,with spinal cord ischemia-reperfusion injury at different time points.To explore whether or not electroacupuncture preconditioning could inhibit the release of the IL-1?,IL-6,TNF-?,NF-?B,TLR4,promote the release of IL-R?,and alleviate the tissue injury and pathological development,then alleviate the spinal cord ischemia reperfusion injury through sympathetic nerve pathway.Method40 New Zealand rabbits were randomly divided into model group,sham-operation group,sympathetic nerve block group,electroacupuncture group and electroacupuncture& sympathetic nerve blocking group,8 rabbits in each.The spinal cord ischemia-reperfusion injury model was established by Zivin method.Sham-operation group: Abdominal operation was performed on rabbits,abdominal aorta wasn't clipped and abdominal suture was performed.Evlectroacupuncture group: Two points of "Xuanzhong" and "Yanglingquan" were treated by electroacupuncture before modeling,at 30 minutes before modeling.Model group: The rabbits were placed on the special platform with supine position.Abdominal cavity was cut open;the abdominal aorta was clipped with the artery clip for 30 minutes,and then removed the artery clip for 30 minutes to recover the blood flow,and then suture the abdomen.No treatment was operated in this group.Sympathetic nerve block group: 30 minutes before modeling,after anesthesia,the rabbit was placed on a special platform with supine position;the right cervical sympathetic nerve was separated,transected,to create sympathetic nerve transection.Electroacupuncture preconditioning & sympathetic nerve block group: 30 minutes before modeling,after anesthesia,the rabbit was placed on a special platform with supine position;the right cervical sympathetic nerve was separated,transected,tocreate sympathetic nerve transection.Then treat the rabbits by electroacupuncture for 30 minutes.2mL blood was collected in each group from femoral vein at four time points:0 h before modeling,4 h,8 h and 12 h after ischemia-reperfusion.After 12 hours of reperfusion,the animals were killed,and then the L2-L5 spinal cords of the rabbits were collected for examination.The content of IL-1?,IL-1?,IL-6,TNF-? in serum was detected by ELISA method,the protein of NF-? B,TLR4 in injured spinal cord was measured by Western blot method,and the expression of IL-6,TNF-? in spinal cord was detected by immunohistochemical method.Result1.Model group: compared with sham-operation group,the content and expression of IL-1 ?,IL-1R?,IL-6,TNF-?,NF-? B,TLR4 increased at all time points(P < 0.05).2.Compared with the model group,the content and expression of IL-1?,TNF-?,NF-?B,TLR4 in the electroacupuncture group were significantly lower than those in the model group at 4 h and 12 h after spinal cord ischemia and reperfusion(P < 0.01),and the level of IL-6 in the EA group was significantly higher than that in the model group at 4 h and 12 h,respectively.However,IL-1R? began to rise at 4 h and began to decrease at 8 h and rise at 12 h.In the electroacupuncture group,the content of IL-1R?was increased by decreasing the content of IL-1?,TNF-?,NF-?B,TLR4,thereby protecting the injured spinal cord tissue.3.Compared with the electroacupuncture sympathetic nerve transection group,the content and expression of IL-1 ?,TNF-?,NF-?B,TLR4 were higher than those in the electroacupuncture group at 4 h and 8 h and 12 h after reperfusion.However,the content of IL-6 was lower at 4h and 8h and higher at 12 h than that in EA group.There was significant difference between the two groups(P < 0.05).4.Compared with the model group,there was no significant difference in the content and protein expression of IL-1?,IL-6,TNF-?,NF-?B,TLR4 between the sympathetic nerve block group and the model group,but there was no significant difference between the model group and the model group(P > 0.05).5.Compared with the model group,the content and protein expression of IL-1?,IL-6,TNF-?,NF-?B,TLR4 in the electroacupuncture & sympathetic nerve block group were lower than those in the model group(P < 0.05),IL-6 increased at 4 h and 8 h,and decreased at 12 h than in the model group.ConclusionThe therapeutic mechanism of electroacupuncture on spinal cord ischemia-rep erfusion injury may be that electroacupuncture could inhibit the level of IL-1?,I L-6,TNF-?,NF-?B,TLR4,promote the expression of IL-1R?,and slow down p athological development and tissue injury,thereby reduce spinal cord ischemia-re perfusion injury.
