Objective: To explore the relationship between DNA repair gene RAD51 D 3'-untranslated region(3'-UTR)mi RNA target-polymorphism and susceptibility to hepatocellular carcinoma(HCC)in guangxi area,to provide the scientific basis for prevention and treatment of HCC.Methods: A hospital based case-control study was used to investigate the relationship between the single nucleotide polymorphisms(SNP)rs12947947 A>G and rs28363292 T>G RAD51 D 3'-UTR mi RNA target polymorphism and the genetic susceptibility with hepatocellular carcinoma.In this study,806 patients with hepatocellular carcinoma and 846 non tumor controls matched from January 2007 to April 2011 in First Affiliated Hospital of guangxi Medical University and Affiliated Tumor Hospital of guangxi Medical University.By using multiple factor Logistic regression statistics,the odds Ratios(OR)and its 95% confidence intervals(95%CI)were calculated for the associations between genotypes of the above two SNPs and susceptibility to HCC.Results: There was no significant difference in the distribution of age and sex between the case group and the control group(P>0.05),and the frequency of smoking,drinking habit and HBV infection in the case group was much higher than that of the control group(P<0.05).In the control group,thedistribution of SNP genotype was in line with the Hardy-weinberg genetic balance(rs12947947 P=0.10,rs28363292 P=0.41).After adjusted by age,sex,smoking,drinking status and HBV infection,it was found that compared with the rs12947947 AA genotype,the rs12947947 AG genotype of the RAD51 D gene significantly increase the risk of individual HCC(OR=1.52,95%CI=1.52(1.00-2.29,P=0.049).Further stratified by age,sex,smoking,drinking status and HBV infection,found that among non smokers,the risk of HCC was significantly increasing in the A genotype carriers than those with the RAD51 D gene rs12947947 GG genotype(AG/AA vs.GG:OR=1.68,95%CI=1.01-2.80,P=0.04).HBV negative population,the risk of HCC significantly increasing among GG genotype carriers than those with the rs28363292 T allele(GT/TT vs.TT:OR=1.76,95%CI=1.06-2.95,P=0.03).Conclusion: The SNPs(rs12947947,rs28363292)of the RAD51 D gene may be associated with the susceptibility to HCC,but the association only in rs12947947 with non-smoker and rs28363292 with HBV negative population.The rs12947947 A alleles carrying genotypes in non-somker are associated with the susceptibility to HCC,and rs28363292 G carrying genotypes genotype in the HBV negative population.This study still needs to be verified by larger samples.
