Chapter 1 Establishing rats active and passive smoking modelObjective:In order to research the effect of active smoking and passive smoking on the body,we constructed the rat's active smoking model and passive smoking model at once,and observed the changes of rats' body weight,oral mucosa,complete blood count and liver and kidney function in active smoking group,passive smoking group and control group,to providing the basis for comparing the influence of active smoking and passive smoking on salivary cytokine of rats.Methods:28 Wistar rats were randomly divided into 3 groups,namely,active smoking group,passive smoking group and control group.We established active smoking model with the device,which was invented by our group.The rats in passive smoking group were exposed to smoking by passive smoking cage.Rats in the control group inhaled the fresh air.Rats in two smoking group smoked for 3 months,twice a day,10 min each time.The rats in the active smoking group and the passive smoking group smoked two times a day,inhaling 2 cigarettes for a period of 3 months.The changes in general view,oral mucosa,body weight,blood cell count and liver and kidney function were observed in rats.Result:The rats in the active smoking group were addicted about 2 weeks.After 3 months of smoking,the oral mucosa of the active smoking group and the passive smoking group showed different clinical changes:the color was pale and the elasticity decreased.By histopathological HE staining,the incidence of white keratosis on the oral mucosa in active smoking group was 75%,and the incidence in passive smoking group was 20%.In the active smoking group,the inflammation of the rats' buccal mucosa were 1-2,in the passive smoking group,the degree of inflammation in the buccal mucosa was 0-1,while in the control group was 0.Compared with the control group,the weight of the rats in the active smoking group and passive smoking group were decreased,the score of the refusal experiment and the suspension experiment were decreased,the time of the slope experiment was prolonged,the red blood cell count,the white blood cell count,the mean corpuscular volume,the percentage of lymphocyte,the count of platelet and the alkaline phosphatase level were significantly increased and the level of creatinine was decreased.At the same time,compared with the passive smoking group,the body weight of the active smoking group was decreased,the score of the suspension experiment was lower,the time of the slope experiment was prolonged,the red blood cell count,the mean corpuscular volume and platelet count were increased significantly,and the creatinine level were decreased.Conclusions:The physical and chemical stimulation of cigarettes will influence the rat's oral mucosa,weight,nerve behavior,whole blood cell count,alkaline phosphatase and creatinine.Chapter 2 Influence of active smoking and passive smoking on salivary IL-6,IL-12 p70 and IFN-y levels and correlation between this cytokine with other parametersObjective:The purpose of this study was to investigate the influence of active smoking and passive smoking on the levels of IL-6,IL-12 p70 and IFN-y in saliva of rats,and the correlation between IL-6,IL-12 p70 and IFN-y with the grading of inflammation in buccal mucosa,gingival index,body weight,whole blood cell count and the liver and kidney function in rat.Methods:After 3 months of experiment,the levels of IL-6,IL-12 p70 and IFN-y in saliva of all three group rats were measured using ProcartaPlex Multiplex Immunoassays combined with Luminex.SPSS 20 was used to analysis the level of salivary cytokine in three groups,and compared the correlation between salivary cytokine with the inflammation grading of the buccal mucosa,gingival index,the weight,the whole blood cell count and the liver and kidney function.Result:Compared with passive smoking group and control group,the levels of IL-6 in the saliva of active smoking group were increased.Compared with the control group,the salivary IL-12 p70 was increased in the active smoking group and passive smoking group,and the difference was statistically significant.However,there was no significant change in IFN-y in the three groups.In addition,the level of IL-6 in saliva of rats was negatively correlated with body weight,creatinine and uric acid.There was also a positive correlation with the inflammation grading of the buccal mucosa,gingival index,white blood cell count,red blood cell count,lymphocyte percentage,platelet count,mean corpuscular volume and alkaline phosphatase.The IL-12 p70 level in saliva was negatively correlated with rat weight and white fine.There was a positive correlation between the inflammation grading of the buccal mucosa,gingival index,white blood cell count,red blood cell count and alkaline phosphatase.Conclusions:Active smoking would induce inflammation in the oral cavity and increase the level of IL-6 and IL-12 p70 in saliva.Passive smoking would also increase the level of IL-12 p70.In addition,IL-6 and IL-12 p70 also had a certain correlation with the inflammation grading of the buccal mucosa,gingival index,body weight,partial whole blood cell count and liver and kidney function in rats.
