Purpose:.In order to find and identify the differently expressed proteins in aqueous humor(AH)of humans between PDR patients and patients with senile cataract.,we uesed the technology of TMT and LC-MS-MS to find and quantify the differently expressed proteins which involve in the pathogenesis of PDR by the analysis of the bioinformatics.Some potential possible biomarkers of patients with PDR may be filtered through this course.Methods:We included twenty subjects in total.10 with PDR(PDR group)and 10 with simple cataract(control group),were involved in our study.The AHs of 20 subjects were then collected under the ophthalmic operating microscope,,and stored at-80? at once.The samples of all the subjects were digested by trypsin,then as a consequence these proteins were divided into some peptides.These peptides were labeled with TMT reagents.Next,we used a Liquid Chromatography-tandem mass spectrometry analysis(LC-MS/MS)to assay the differently expressed proteins and evaluated them by relevant databases and manual analysis according to the pre-planned standard.After that,we manufactuered a bioinformatics analysis to search the role and relationship of these selected proteins and possible biomarkers of patients of PDR.could be forecastedResults:According to our quantitative proteomics analysis,we verified 319 human proteins in all.127 proteins of which were believed to be significant,since they just existed in all the 20 samples.The differently expressed proteins with up-regulating greater than 1 times and p-value less than 0.05 were 28 proteins,and the differently expressed proteins with down-regulating less than 1 times and p-value less than 0.05 were 16 proteins.We concluded that FIB?a2-MG?AGT?C3?C4?APOAl and Clstn-1 may play an important role in the pathogenesy of PDR.Conclusion:Through this research,we created a database of AH differently expressed proteins of PDR patients,and verified some significant proteins triumphantly.According to the bioinformatics analysis,we discovered that FIB??2-MG?AGT?C3?C4?APOAl and Clstn-1 may be the possible biomarkers of PDR.Researchers concerned may investigate the pathogenesis of PDR further by these proteins mentioned obove in the near future.
