Objective:To explore the association between SNPs of PTGS2 gene promoter region and clinicopathologic characteristics and prognosis to HCC after resection.Methods:The genotypes of the-1195G>A,-765G>C polymorphisms of PTGS2 gene promoter region were determine by polymerase chain reaction-restriction fragment length polymorphism(PCR-RFLP)analysis using DNA from Peripheral blood of with initial,undergoing RO resection.For each polymorphism,the Hardy-Weinberg equilibrium was tested,?2 test was used to calculate the correlation between the genotypes and patient clinical characteristics.Survival distributions were estimated with the Kaplan-Meier method and were compared with the log-rank test.Multivariate Cox proportional hazards models were applied to estimate the effect of prognostic factors on OS and DFS.All statistical tests were two sided,and P<0.05 were used as a criterion of significance..Results:Two hundred patients,all observed genotype frequencies of PTGS2 gene-1195G>A and-765G>C in patients conformed to the Hardy-Weinberg equilibrium(P>0.05).No statistical associations were observed between any of these polymorphisms and sex,age,Child-Pugh grade of liver function,preoperative serum AFP level,Primary tumor size,number of tumor mass,portal vein invasion status,histologic classification and clinical stages.The DFS for patients with the-1195AG genotype was shorter(P= 0.005 for log-rank test)than those with the-1195GG genotypes.Moreover,the DFS for patients with the-1195A allele genotypes(AA + AG)was also significantly shorter than that for patients with the GG genotype(P = 0.008 for log-rank test)by Kaplan-Meier estimates.Patients that in the clinical stages ? with the-1195A allele(AA+AG)was also significantly shorter than that for patients with the GG genotype(P=0.024 for log-rank test).No significant associations were found among other PTGS2 polymorphisms and clinical outcomes(all P>0.05 for log-rank test).The multivariate analysis revealed that for DFS,preoperative serum AFP level?400ng/ml(P=0.006,HR:0.495,95%CI:0.298-0.820)was independent factor;for OS,preoperative serum AFP level?400ng/ml(P=0.001,HR:0.388,95%CI:0.222-0.679)and the moderate or high differentiation(P=0.022,RR:0.387,95%CI:0.171-0.872)were independent factors;-1195G>A and-765G>C polymorphisms were not independent ly associated with DFS and OS.Conclusion:No statistical associations were observed between-1195G>A and-765G>C polymorphisms of PTGS2 gene and clinicopathologic characteristics.and-1195AG genotype and A allele were significantly associated with patients DFS.Moreover,-1195G>A polymorphism were significantly associated with OS of patients that in the clinical stages ?,and-765G>C polymorphisms had no associations with DFS and OS of HCC.-1195G>A and-765G>C polymorphisms were not independent ly associated with DFS and OS.
