Polygoni Multiflori Radix,the dry root of Polygonum multiflora Thunb.,named Heshouwu in China,is commonly used as traditional Chinese medicine,in raw and processed dual-use system.With warm qualities and bitter and sweet flavor,the raw is good at removing toxicity,eliminating carbuncles,preventing malaria and relaxing bowel.While the processed one becomes warmer and sweeter,with the functions of invigorating liver and kidney,benefiting life essence and nourishing blood,strengthening muscle and bone,making hair dark and reducing lipid.The changes on flavors and functions lead to significant differences in clinical application,which is closely related to the effect of the processing on the chemical material basis of Polygoni Multiflori Radix.The processed products of Polygonum multiflorum are widely used in clinical application.However,the processed Heshouwu in the market is vary in quality at present,and the reason is that there is no uniform standard for the processing technology.Although the processing method of Heshouwu was collected in The 2015 edition of"Chinese Pharmacopoeia",The key processing parameters are not specified.The relationship between the dual-use principle and its intrinsic material base is still to be analyzed deeply.The relationship between the application of the raw Heshouwu and processed products with its intrinsic material basis has to be analyzed deeply.ObjectiveTo improve the quality and to provide the scientific basis for the traditional processing theory of the Heshouwu based on the analysis of the main functional components of Heshouwu by the orthogonal optimization.And the changes of various components in the process of processing and its variation patterns were investigated as comprehensive as possible.So as to analyze the s c i e n t i f i c c o n n o t a t i o n o f d i f f e r e n t a p p l i c a t i o n s y m p t o m s o f unprocessed and steamed Heshouwu from the changes and differences of chemical composition.To provide some evidence for clinical practice,promote the r a t i o n a l u s e o f m e d i c i n e a n d p r e v e n t t h e h a r m o f t h e m i x i n g.Methods1.Eight groups of Polygoni multiflori Radix were processed.Then the contents of polysaccharides,stilbene glycoside,anthraquinone,lecithins and ethanol-soluble extractives from different regions and different growth stages were compared to evaluate the quality of Heshouwu.2.Heshouwu grow in Guizhou province for three years was selected as a sample for processing.The effect of the steaming time,drying time,and drying temperature were investigated by taking polysaccharides and alcohol extract as the indexes in single-factor test.Through L9(33)orthogonal test design,the process on heshouwu of black bean juice was optimized by measuring the content of polysaccharide,stilbene glycoside,anthraquinone,lecithin and ethanol-soluble extractives And select the best processing technology.The quality standard of processing with black bean juice may be established by comparing the index content of the guizhou three-year-old and annual Guangdong heshouwwu.3.The orthogonal experiment of L9(33)was used to optimize the process of steaming with water,and the content of polysaccharide,stilbene glycoside,anthraquinone,lecithin and alcohol extract in the process of steaming polygoni multiflori Radix were studied,and the best processing technology would be optimized.4.Compare the contents of raw materials,black soya bean juice system,water steamed system of processing technology,and compare the indications effect between them,reveal the processing principle,to determine the optimum degree of processing changes in medicine to establish a scientific and reliable Polygonum multiflorum Pieces quality evaluation system.Results1.According to the established method,the contents(polysaccharides,stilbene glycoside,anthraquinone,lecithin and ethanol-soluble extractives)of 8 batches of Heshouwu in different producing areas,regions and growing years were determined.From the point of view of polysaccharides and lecithins,Guizhou Meitan>Guizhou Shibing>Guangdong,three-year-old is better than one-year;from the perspective of stilbene glycoside,anthraquinone,Guangdong>Guizhou Meitan>Guizhou Shibing,one-year-old is better than three-years-old.2.The optimal processing technology as follows were determined by the comprehensive scoring method,the processing time was 8 h×3 days,the drying temperature was 80°C,and the drying time was 8 h.Other processing parameters were moisturizing 12 hours with Heshouwu for 1:10,while steamed with pure water for 1:1 moisturizing for 12 hours.Orthogonal experiments show that the concoction conditions influence the processing effect as follows:processing time>drying temperature>drying time.According to the optimized concocting process amplification test,the results showed that the quality of steamed and black bean juice system was higher than the pharmacopoeia standard;compared with the orthogonal test,all the detection indicators tend to the upper limit,and more stable.3.The HPLC method was used to analyse the contents of stilbene glycoside,free anthraquinone,and combined anthraquinone in raw and processed Heshouwu.The content of polysaccharides and lecithins were determined by uv-vis spectrophotometry.The results showed that during processing,the contents of stilbene glycoside,lecithin,and polysaccharides decreased;the contents of free anthraquinone increased,and the content of combined anthraquinone decreased significantly.The combined anthraquinone may stimulate large intestine mucosa,which is closely related to the side effects of Heshouwu.In processing,the anthraquinones gradually shifted from combined state to the free state,which can reduce its toxic and side effects on the human body and enable it to exert its physiological activity.ConclusionThe processing technology of Polygoni Multiflori Radix optimized by orthogonal experiment is superior,stable and repeatable.In the processing process,the effective ingredients in Polygonum multiflorum as stilbene glycosides,lecithin,polysaccharides and so will inevitably be a loss,we should pay attention to its maximum retention;try to make combined anthraquinone turning to the free state,even to a maximum,in order to achieve the purpose of increasing efficiency and attenuation.The effective content of the products show significant difference,clinical application should be distinguished based on their specific taste and efficacy.
