Purposelingnan region with southeast Asia monsoon maritime climate characteristics,high temperature and humid environment as the main climatic characteristics.Diet of high protein relate to deficiency of spleen and stomach,combined with the lingnan area unique natural climate,the fibrile disease Danmp-Heat Syndrome is common in patients with the heat and humid factors and spleen as the center and the internal injuries.Is the basis and internal cause of the disease,hot and humid factors is one of the key point links it's model replication.using the automatic artificial climate cabinet to maintain temperature and humidity conditions.To establish models of damp-heat syndrome rats by compound factors to establish the mouse model serving for Fibrile Disease Damp-Heat Syndrome,this aim is to explore composition of intestinal flora,inflammatory status and the change of aquaporins in the Febrile Disease Danmp-Heat Syndrome.MethodsUsing hight protein diet with humidity and heat environment and other biological compound factors,to establish the mouse model of Damp-heat syndrome.Wet factors totally enclosed automatic temperature control of artificial climate box raise the temperature and humidity.To mantain lingnan unique thermal and humid environment,using intraperitoneal injection of LPS simulation to rage invasion wet as the characteristic.We establish the damp-heat model to monitor daily activities and signs,measuring mice body weight changes every week,using HE staining to detect tongue pathomorphism changes;Serum LPS THF-? concentration was detected by enzyme-linked immunosorbent assay(ELISA).Immunohistochemical method was used to detect the protein positive expression of NF-kb p65 TLR4 AQP3 and AQP4 in mouse model colon tissues.ResultsSymptoms of damp heat syndrome of damp syndrome model were manifested in the symptoms of weight loss,sleeping laziness,anorexia and sticky perianal filth of the stool.Colon pathology results show that the thickness of tongue coating of dampness-heat syndrome with predominant dampness group is significantly higher than dampness-heat syndrome with predominant heat group;the four kinds of mouse models content is highter than the normal group,the serum level of LPS,THF-?,NF-kb p65,TLR4 in dampness-heat syndrome with predominant heat group increased significantly compared with normal group(P < 0.001),compared with group LPS mouse model,the level of LPS?TNF-? in dampness-heat syndrome with predominant heat group is greater than the dampness-heat syndrome with predominant danmp group(P < 0.05),the NF-kbp65 and TLR4 protein expression in two kinds of damp-heat syndrome mouse model had no significant difference;Compared with the dampness-heat syndrome with predominant damp group,the expression of AQP3 and AQP4 increased in the dampness-heat syndrome with predominant damp group(P < 0.05).It was suggested that the inflammation degree in the damp group was higher than that of the heat group.It was presumed that it may be related to the influence of alcohol on the permeability of the intestine and the damp and hot environment,and the inflammatory factors and the aquaporin.Protein disorder may be one of the quantitative criteria in animal models of damp heat syndrome.Test report of manure 16 srRNA show that the dominant bacteria group in two groups of damp heat syndrome animal is Bacteroidetes and Firmicutes;results of PCA microbial communities are similar,the diversity of Alpha results suggest that the two heat model group richness decreased.ConclusionAccording to the modeling method of the syndrome of damp heat syndrome of traditional Chinese medicine,two mouse models of damp heat syndrome were established by the composite factor modeling method.After the model was completed,the mice in the model group showed similar signs of clinical damp heat syndrome and symptoms;Intestinal microflora results showed that the intestinal microflora in the mice of the two groups of damp-heat syndromes with mild illnesses decreased,and the changes in Bacteroidetes,Firmicutes,and Clostridia may be the characteristics of moist heat in mice with damp-heat syndromes;animal models of febrile illness syndrome exist inflammatory mediators induced by the endotoxin-TLR4 pathway and disturbances of aquaporins,which may play an important role in the quantification of damp and heat,and in the determination of damp and hot stress.
