Effect of curcumol on the regulation mechanism of inositol dependent enzyme 1(IRE1)pathway in renal tissue of rats with unilateral ureteral obstruction(UUO).Animal models of renal interstitial fibrosis were established by unilateral ureteral obstruction.According to the experimental requirements,a certain amount of rats were selected and divided into low dose group,medium dose group,sham operation group and high dose curcumol group for comparative study.After operation,rats were sacrificed and serum and kidney tissues were collected.Of serum creatinine(Scr),urea nitrogen(BUN)related indicators were detected.The left kidney was harvested and the renal edema level was analyzed.HE staining was used to determine the renal pathological changes.The protein expression of IRE1 and TRAF2 in renal tissues was detected by protein embolism.Apoptosis of renal tubular epithelial cells index.?Serum levels:on the 14th day after operation,Scr and BUN in the model group were significantly higher than those in the sham operation group(P<0.01);Scr(P<0.05).Compared with enalapril,the high dose of Curcumol decreased the expression of Scr and BUN obviously.?The degree of renal interstitial fibrosis:the increase of relative area of renal interstitial collagen in model group(P<0.01).The relative area of renal interstitial collagen in each treatment group decreased significantly,which was significantly different from that in model group.Compared with enalapril group,The relative area of interstitial collagen in the renal interstitium decreases(P<0.05).?The index of renal tubular injury:The degree of renal tubular injury in model group was significantly higher than that in sham operation group(P<0.01)Compared with the model group,the injury of renal tubule was obviously alleviated.Compared with the enalapril group,the high dose of curcumol had a better remission(P<0.05).?Compared with the model group,kidney weight increased significantly,knot(P<0.01).The renal weight of each treatment group decreased to a certain extent,which was significantly different from that of the model group.Compared with the enalapril group,the curcumol high-dose group had a greater reduction of renal weight(P<0.01).?Compared with the model group,the protein expression of IRE1 and TRAF2 in each treatment group was significantly higher than that in the sham-operated group(P<0.05,P<0.01).Compared with the enalapril group,the Curcumol high-dose group could significantly reduce the expression of IRE1 and TRAF2 protein(P<0.05);?The apoptosis of renal tubular epithelial cells The apoptotic tubular epithelial cells in model group were significantly higher than those in sham operation group(P<0.01),and the apoptosis index in each treatment group was significantly decreased compared with model group.Curcumol high dose Compared with enalapril group,apoptotic index decreased significantly(P<0.05).Curcumol can protect renal function in rats with unilateral ureteral obstruction;renal lesions alleviate hydronephrosis;reduce UUO interstitial fibrosis in a rat model of renal tubular epithelial cells,vacuolar degeneration,interstitial infiltration,renal tubular atrophy,red blood cell casts,interstitial edema and protein tube,renal tubular expansion,thereby reducing the tubulointerstitial injury;reduce kidney interstitial collagen fibers in rats,the excessive deposition of extracellular matrix has improved,hinder the development of renal fibrosis;Decrease the expression of IRE1,thus the apoptosis pathway mediated by IRE1-TRAF2 interference pathway,and prevent renal interstitial fibrosis caused by renal tubular epithelial cell apoptosis.
