Objective:To investigate the effect of HOTAIR on proliferation,migration and invasion of human tongue squamous cell carcinoma cell line.Methods:The expression of HOTAIR was knocked down by liposome mediated siRNA,and the meaningless sequence was set as the control group.QRT-PCR was used to detect the silencing effect.MTT assay was used to detect the proliferation of 2 tongue squamous cell carcinoma cells,the changes of invasion and migration ability of 2 tongue squamous cell carcinoma cells were detected by scratch test and Transwell test.Results:In the 2 cell lines,HOTAIR was successfully knocked down and the knockdown efficiency was 93.13%?P<0.0001?and69.28%?P<0.0001?,respectively.The absorbance of cells were detected at different time points:in addition to 0h,the absorbance of cells in the experimental group was significantly lower than the control group(P0h>0.05,the rest of the time points P<0.05),proved that the cell proliferation was inhibited after the fall;In MTT assay,the membrane cell number of the experimental group of the cell lines was significantly less than the control group?P<0.0001?,the migration distance of the experimental group in scratch group was less than the control group?P<0.0001?,proved that the invasion and migration ability of cells decreased after knocking down.Conclusion:Long non-coding RNA HOTAIR promotes the tongue squamous cells carcinoma?Tca-8113,TSCCA?on the proliferation,migration and invasion,and involved in the development of tongue cancer. |
