Objective:To investigate the effects of long-chain non-coding RNA HOTAIR(lnc RNA HOTAIR)on the expression of Notch signaling pathway and EMT in tongue carcinoma cells Tca8113 and TSCCA.Methods:Liposomes-mediated small interfering RNA(siRNA)was used to down-regulate the expression of HOTAIR in tongue carcinoma cell lines TCA8113 and TSCCA,and was divided into HOTAIR down-regulated group and control group.Real-time fluorescent quantitative PCR was used to detect down-regulation.The effect of Western blotting was used to detect the expression of Notch signaling pathway and EMT-related proteins.Results : The HOTAIR was successfully down-regulated in the tongue carcinoma cell lines Tca-8113 and TSCCA.The difference was statistically significant(P<0.05).In the two cell lines,the results of Western blotting showed:(1)The Notch pathway in the HOTAIR down-regulated group was affected.The protein expression of Notch1,ligand Jagged1 and downstream target gene HES1 was significantly lower than that of the control group,and the difference was statistically significant(P<0.05).(2)Compared with the control group,the EMT-related protein in the HOTAIR down-regulated group the protein expression of E-cadherin was up-regulated(P < 0.005),Vimentin protein expression was significantly decreased(P <0.005).The difference was statistically significant.Conclusion:HOTAIR is involved in the regulation of Notch signaling pathway and promoting EMT in tongue carcinoma cells Tca-8113 and TSCCA,which may be an important meeting point for the regulation of EMT in tongue cancer Notch signaling pathway.
