ObjectiveThe rat model was established by subcutaneous injection of calcium gluconate in rats.To study the effects of aloe combined with HouFeng-san on the damage of calcium extravasation,through animal experimentation.MethodsBuilding animal models:The 16 rats were randomly divided into four groups:blank group,low dose group?0.5mL?,medium dose group?1mL?and high dose group?1.5mL?.The blank group does nothing.The other groups were used to build animal models by subcutaneous injecting of calcium gluconate on the back.To selecte the appropriate dose by comparing the degree of extravasation and histopathological changes.Intervention experiment:The 40 rats were randomly divided into blank group,model group,magnesium sulfate group,aloe group and aloe-HouFeng-san group.Andthenestablishedmodelofextravasationofcalcium gluconate.According to the group,the physiological saline,magnesium sulfate,aloe gel and aloe gel throat powder were used.According to the group,we used saline,magnesium sulfate,aloe gel and aloe-HouFeng-san in 30 minutes after molding.And the blank group does not intervene.Recorded the changes in the wound surface and took photos forcalculating the damage area after the intervention 1,4,7,11,18 days.Each group executed one rat at a time point.Wound tissue pathology HE staining was used to observe the histopathological changes,tissue homogenatewas used to detecte tumor necrosis factor alpha?TNF-??and basic fibroblast growth factor?bFGF?concentration by the way of enzyme linked immunosorbent method?ELISA?.Statistical software SPSS19.0 was used for statistical analysis.Results1.Successfully established the rat model of calcium gluconate extravasation.2.Therewasnopathologicalchangeintheblankgroup.The histopathological changes and the degree of extravasation of aloe-HouFeng-san group were less than the model group,magnesium sulfate group and aloe group.The damage area of aloe-HouFeng-san group was smaller than other intervention groups.The concentration of TNF-?and bFGF in each group was increased and then decreased after the extravasation.At 1stday after intervention,the concentration of TNF-?in aloe-HouFeng-san group was higher than other intervention groups,and the difference was statistically significant?P<0.05?.At 4th,11thand 18thday after intervention,the concentration of TNF-?in aloe-HouFeng-san group was lower than other intervention groups,and the difference was statistically significant?P<0.05?.The bFGF in aloe-HouFeng-san group was higher than other intervention groups,and there were significant difference at 4th,7thand 11thday after intervention?P<0.05?,there was no statistically significant difference at1thand 18thday after intervention?P>0.05?.There was no significant difference in TNF-?and bFGF concentration between magnesium sulfate group and aloe group at all time points?P>0.05?.ConclusionThe effect of Aloe-HouFeng-san on the wound healing of calcium extravasation is better than magnesium sulfate and aloe.Aloe-HouFeng-san can reducetheinflammatoryinfiltration,promotetheproliferationof fibroblasts,reduce the necrosis of the skin and promote wound healing for calciumextravasationinjury.Intheearlystageofwound healing,aloe-HouFeng-san can stimulate the release of TNF-?to promote inflammatory response.And it can inhibit the excessive release of TNF-?to avoid tissue damage at the later stage of wound healing.Aloe-HouFeng-san can stimulate the release of bFGF early and keep bFGF at a high level to promote wound healing and reduce scar.
