Research On The Effect Of Acupuncture And Moxibustion On Interleukin-4(IL-4) And Interferon-gamma(IFN-?) In Serum And Small Intestine Tissue Of Mice With Cyclophosphamide

Obgective This study with Cyclophosphamide(CTX)to immune injury in mice as the breakthrough point,through the observation of acupuncture before and after the intervention of thymus and spleen index and the small intestine in mice's junction(PP)number?morphology,and the mice serum and intestinal tissue of interleukin 4(IL-4),interferon gamma(IFN-?)and the ratio of IFN-?/IL-4 change,from the intestinal immune system and the overall correlation and difference and immune balance,etc.to reveal the mechanism of the acupuncture and moxibustion improve immune injury caused by CTX.Methods Eighty SPF male kunming mice were selected and purchased in two batches,40 in each batch.The two batches were: group 3d and group 5d respectively.Each batch was divided into four groups:blank group,model group,acupuncture group and moxibustion group,eatch group have 10 mice.In the blank group,normal saline was injected into the abdominal cavity for 100mg/kg/d according to the weight of the mice,for 3 consecutive days.The remaining 3 groups of mice were given the same dose of CTX according to the same method.Four groups of mice to select the same number in mice began to fixed at the same time,in addition the needles,the moxibustion group selection “Dazhui”,“Geshu”,“Shenshu”,“Zusanli” were given acupuncture and moxibustion therapeutic intervention,3d and 5d group mice intervention in 3 consecutive or five days,respectively.Groups of mice respectively at the end of the treatment after 24 h after eyeball blood,centrifugal,take on in-20 ? refrigerator for inspection,Anatomic mice were quickly removed from thymus,spleen,small intestine,and weighed thymus and spleen.The number and size of small intestinal pineal junction were observed with the naked eye.Interception of small intestine jejunum segment in-80? refrigerator storage for testing;The contents of IL-4 and ifn-renewal in serum and small intestinal tissues of mice were measured by ELISA,and the ratio of IFN-?/IL-4 was calculated.The data obtained were input by SPSS 20.0 for Windows software,and the results were analyzed.Results 1.The changes of thymus and spleen index were the same in the 3d and 5d groups.After CTX injection,the indexes of all groups of chemotherapy decreased(P<0.05);after acupuncture and moxibustion,the indexes of both acupuncture and moxibustion groups were increased(P<0.05);the effect of acupuncture was comparable to moxibustion(P>0.05).2.Serum IL-4,IFN-?,and IFN-?/IL-4 levels in 3d and 5d groups showed that the immune cytokines IL-4 and IFN-? decreased in each group after chemotherapy(P<0.05).The ratio of IFN-?/IL-4 decreased(P<0.05).After acupuncture and moxibustion treatment,serum IL-4 and IFN-? increased in acupuncture and moxibustion groups(P<0.05).And the ratio of IFN-?/IL-4 was increased(P<0.05);Compared with the acupuncture group,the levels of IL-4 and IFN-? in the serum of the moxibustion group were more(P<0.05),and the 3d group of moxibustion group The ratio of IFN-?/IL-4 was larger(P<0.05),and the moxibustion group had no significant increase in the 5d group(P>0.05).3.IL-4,IFN-? and IFN-?/IL-4 in small intestine of 3d group and 5d group showed:after injection of CTX,the secretion of IL-4 and IFN-? in the small intestine of each group of chemotherapy decreased(P<0.05).The ratio of IFN-?/IL-4 in the small intestine of the model group decreased(P<0.05).After acupuncture and moxibustion treatment,the secretion of IL-4 and IFN-? in the small intestine increased in the acupuncture and moxibustion groups(P<0.05),and the ratio of IFN-?/IL-4 increased(P<0.05);Compared with the acupuncture group,the secretion of IL-4 and IFN-? in the small intestine of the moxibustion group was less(P<0.05).There was no significant difference in the ratio of IFN-?/IL-4 between the treatment and moxibustion groups(P>0.05).Conclusion1.Acupuncture can increase the thymus and spleen index of mice and repair the damage of immune organs caused by immunosuppression.2.Acupuncture can increase the levels of immune factors IL-4 and IFN-? in serum,regulate the suppressed immune function back to normal,and reduce the overall immune damage caused by chemotherapy.At the same time,acupuncture can up-regulate the ratio of IFN-?/IL-4 in serum,make the relative increase of IFN-? in serum and decrease the production of IL-4,the balance of immune balance recovers,the tendency of immune response is Th1,the balance of Th1/Th2 tends to be stable.3.Acupuncture can promote the secretion of immune factors IL-4 and IFN-? in the small intestine,repair the damaged intestinal immune function,and reduce the intestinal mucosal immune damage caused by chemotherapy.At the same time,acupuncture can upregulate the ratio of IFN-?/IL-4 in the small intestine,increase the secretion of IFN-? in the intestine,decrease the secretion of IL-4,restore the balance of the immune balance in the intestine,and increase the immune response in the intestine Th1.In the intestine Th1/Th2 balance tends to be stable.
