Objectives1.To study the effect of Jasminum extract on ulcerative colitis mice;2.To investigate the effect and mechanism of Jasminoside on inflammatory reaction in LPS-induced human colonic epithelial CRL-1790 cells.Methods1.Animal experiments in vivo: use TNBS enema to establish mice models of ulcerative colitis.The mice were randomly divided into normal control group,model group,positive control group and experimental group,.The experimental group was treated with Jasminum extract,the positive control group was treated with sulfasalazine,the model group and the normal control group were treated with the same amount of saline.After drugs intervention,the behavioral performances of mice were observed.After animals sacrificed,the colon morphological changes were observed,and the levels of colon SOD,MPO and MDA in colon tissues were detected by using the kits.2.Cell experiments in vitro: the activity of human colon epithelial cells was assayed by MTT assay to select the experimental concentrations.Cells were pre-incubated with different experimental concentrations of Jasminoside and then incubated with LPS to induced cell inflammatory response.Using ELISA to detect the cytokines levels of IL-1,IL-8 and TNF-?.Using PCR to detect the m RNA expression of CYP3A4 and CYP1A2.And using Western blot to detect the protein expression of NF-k B p65 and PXR.Results1.Animal experiments in vivo: compared with the model group,experimental group obviously improved symptoms,significantly reduced colon tissue injury,significantly increased the level of SOD but decreased MPO and MDA.2.Cell experiments in vitro: Jasminoside concentrations of 10 ?mol/L,20 ?mol/L and 40?mol/L had no effect on the activity of cells,which were selected as the experimental concentrations.Compared with the control group,LPS significantly induced the secretion of IL-8 by cells,but had no significant effect on that of TNF-? and IL-1.Compared with LPS-induced group,Jasminoside significantly inhibited the secretion of IL-8 and protein expression of NF-k B p65,but increased the protein expression of PXR and the m RNA relative expression of CYP3A4 and CYP1A2.Conclusions1.Jasminum extract can significantly improve the condition of mice with ulcerative colitis.2.Jasminoside can inhibit LPS-induced inflammatory response in human colonic epithelial CRL-1790 cells.Its possible mechanism is upregulating the expression of PXR to upregulate the expression of downstream metabolic enzymes CYP3A4 and CYP1A2 and inhibit the expression of NF-k B p65,and ultimately to inhibit IL-8 secretion.
