Modified Tongfu Decoction On The Expression Of Colonic Epithelial Cell Apoptosis In Rat Model Of Ulcerative Colitis

Purpose：This study was to observe and explore the jia wei tong fu souptherapeutic effect in ulcerative colitis(UC) by discussing the influences ofjia wei tong fu soup in rat model of UC on colon tissue epithelial cell apoptosisand protein expressions of regulatory genes such as Bcl-2as well as Bax, andhoped to be able to provide a sufficient experimental evidence in Chinesemedicine treatment of ulcerative colitis.Material and method：70SpragueDawley(SD)ratsofcleangradeweredividedinto blank control group, model group, fill spleen group、Sulfasalazine group、the high, middle, and low dose of jia wei tong fu soup with completely randomgrouping methods.5%TNBS concentrate and anhydrous ethanol according to thevolume of1:1were equipped into a20mg/kg liquid to establish the rat modelof UC. Since the fourth day after establishing, the normal group and model groupwere filled into stomach with normal saline, while other groups were given fillspleen group、Sulfasalazine group、the high,middle,low dose of jia wei tong fusoup, respectively. All were continuously feeding and then were killed in the21th day. Apply with the TUNEL staining method to detect colon epithelial cellsapoptosis of UC model rats and use SABC method to detect the protein expressionof apoptosis regulatory gene Bcl-2and Bax.Results：Compared with blank control group, the AI value, and proteinexpressions of Bcl-2and Bax significantly increased in the model group, andthere were significant differences via statistical analysis (P <0.01); Comparedwith fill spleen group, the middle and low dose of jia wei tong fu soup, thehigh dose of jia wei tong fu soup possessed the lower AI value, the higherexpression of Bcl-2protein expression as well as the lower Bax proteinexpression, and there were statistical significance (P<0.01); Compared withSulfasalazine group, AI value declined obviously and Bax protein expressionsignificantly decreased in the high dose of jia wei tong fu soup (P <0.01), although the Bcl-2protein expression had no obvious discrepancy (P>0.05).Conclusion：jiaweitongfusoupcanreducethecolonepithelialcellapoptosisof the rat model of UC by adjusting the Bcl-2and Bax protein expressions toachieve therapeutic effect, and the high dose of jia wei tong fu soup had thebest therapeutic effect.
