This dissertation consists of five chapters.The first chapter review the research progress of the secondary metabolites and biological activities of deep-sea fungi in the past10 years.The second chapter describes the chemical composition and biological activity of a Penicillium sumatrense derived from a deep-sea source.In the third chapter,the chemical constituents and biological activities of a Penicillium biourgeianum derived from a deep-sea source are introduced.In the fourth chapter introduces the chemical composition and biological activity of Chaetomium globosum,a deep-sea-derived Chaetomium.the fifth is a summary.We use various spectroscopy analysis methods and techniques,including IR?infrared spectra?;UV?ultra violet spectra?;NMR?Nuclear Magnetic Resonance?;MS?mass spectrum?,and CD?circular dichroism?;Mosher's method etc.31 compounds were identified from the above three strains,Pinicillium sumatrense,Pinicillium biourgeianum,Chaetomium globosum,including 7 structurally new compounds and 24 known compounds.The main structural types of compounds include macrolides?compounds 1-6?,mycophenolic acids?compounds 7-13?,alkaloids?compounds 15-23?,and polyketides?compounds 26-31?.Compounds 1-4,7,and 15-16 are new compounds.We performed different types of bioactivity tests on different types of compounds.We performed bioactivity assays for the inhibition of NO production by macrolide compounds,compound6 has significant activity in inhibiting LPS-induced macrophages RAW264.7 NO production(IC50 0.91?M,stronger than the positive control).We conducted experiments on the biological activity of the mycophenolic compounds against the dimorphic switch of S.scitamineum compounds 8-10,12 and 13 can significantly inhibit the S.scitamineum fungus transforms from the yeast state to the mycelium state,but does not directly kill the fungus.Among them,compound 8 and 9 had the best activity,and the minimum effective concentrations for inhibiting dimorphism of S.scitamineum were 0.94?M and 1.2?m.Respectively,we performed cytotoxic biological activity experiments on alkaloids,among them,compound 20 had stronger cells for LNCaP and B16F10 cells.The venom activity has IC50 values of 0.62?M and 2.78?M.In this paper,the secondary metabolites and biological activities of three deep sea-derived fungi were studied.Seven new structural compounds were isolated and identified from them,and some of them showed strong biological activity.The results of the study indicate the diversity of chemical structures and biological activities of marine natural products,providing a scientific basis for the rational development and utilization of marine biological resources.
