Objectives The model of periodontitis was established by ligating the right maxillary first molar of SD rats with ligation of steel wire and high glucose diet.The animals of periodontitis model were treated with simvastatin,The expression of transforming growth factor-?,interleukin-1 and tumor necrosis factor-? in periodontal tissues was detected by immunohistochemistry after drug treatment,and the effects of simvastatin on inflammation and bone remodeling in periodontitis were investigated.To provide experimental basis for clinical treatment of periodontitis patients.Methods 60 healthy male SD rats of 2 months of age,weighing 220±20 g,were given clean grade,common feed,free drinking water and feeding.After adaptive feeding,the first chapter divided 30 animals into three groups by simple randomization grouping method.10 rats in each group.Experimental group M1: ligated teeth,high-sugar diet.Negative control group M2: no ligation of teeth,high-sugar diet.Blank control group M3: no ligation of teeth,normal diet;The experimental period is 8 weeks.The changes in body weight were monitored weekly throughout the experiment cycle.At the end of the 8th week,the depth of periodontal pocket,gingival index,mobility degree and alveolar bone resorption of the right maxillary first molar four common clinical indicators of periodontitis were measured.After the animals were killed at the end of 8 weeks and their teeth and periodontal tissues were sliced for histopathological observation.In Chapter 2,30 SD rats were randomly divided into three groups after establishing periodontitis model according to the method in Chapter 1,N1 group: simvastatin group,N2 group: normal saline group,N3 group: blank control group.10 rats in each group were kept at constant temperature.The animals were killed at the end of the 8th week,and the teeth and periodontal tissues of the right maxillary first molar were cut off.The expressions of transforming growth factor-?,interleukin-1 and tumor necrosis factor-? in the periodontal tissues were observed by immunohistochemistry.The average optical density of the positive expression region of three factors was analyzed by IPP6.0 software,the results were statistically analyzed by SPSS21.0 software.Results 1 The weight of M2?M3 control group increased continuously,while that of M1 experimental group decreased first and then increased.Compared with the two control groups,the change of body weight was significantly different(P<0.05).2 The index scores of gingival index,tooth mobility and alveolar bone resorption in M2?M3 control group were not changed compared with normal healthy rats,the three indexes of M1 experimental group were all changed,and compared with the control group of M2?M3,the depth of periodontal bag in M1 experimental group was deeper than that in control group,the difference was statistically significant(P<0.05).3 The results of HE staining in the periodontal tissue of the right maxillary first molar in rats: M1 experimental group showed reticular hyperplasia from epithelium to connective tissue,and inflammatory cells infiltrated around it,vasodilation and congestion,hydrolysis of periodontal collagen fibers,alveolar bone resorption was loosely reticulated,which was in accordance with the pathological diagnostic criteria of periodontitis;While that of M2 and M3 control group was in accordance with the normal periodontal histological standard.4 After simvastatin treatment,the immunohistochemical results of periodontal tissue in the right maxillary first molar of rats showed that: The expression of transforming growth factor-? in the experimental group was higher than that of N2 and N3,while the expression of IL-1 and tumor necrosis factor-? was lower than that of control group.Conclusions 1 the experiment proved that SD rat periodontitis animal model could be successfully established by wire ligation and high glucose diet for 8 weeks.2.The experiment proved that simvastatin could promote the expression of transforming growth factor-? in periodontal tissue.The expression of interleukin-1 and tumor necrosis factor-? was inhibited at the same time.
