Objective In our study,we aims to find the association between serum levels of HMGB1(High mobility group box 1)and clinical features of AS patients before and during treatment.Methods Serum HMGB1 was detected in 147 AS patients and 61 healthy controls using ELISA.We evaluated the association between HMGB1 and disease severity indices as well as extra-articular manifestations.Among these AS patients,41 patients were followed-up at 1,3,and 6 months after treatment.Results The serum HMGB1 of AS patients was significantly elevated compared to healthy controls and positively correlated with BASDAI,BASFI,ASDAS-ESR,ASDASCRP,ESR,and CRP,but not with HLA-B27,anterior uveitis,and recurrent diarrhea.We observed that serum HMGB1 paralleled disease activity after treatment.Conclusion Serum level of HMGB1 is higher in AS patients,and to a certain degree,HMGB1 can reflect the activity of AS and be used as a laboratory indicator to reflect the therapeutic reaction.Objective This study aims to find a more convenient strategy of Gray-scale ultrasound to reflect the disease activity and investigate correlation between the Gray-scale ultrasound score and plasma levels of inflammatory cytokines in patients with Rheumatoid Arthritis(RA).Method 62 RA patients and 10 healthy controls were enrolled in this study,and and the plasma levels of IL6,TNF-?,IL-17 A,IL-1? and IL-1? were detected with CBA method.We analyzed the correlation between the above cytokines and DAS28 of these patients.Wherein 38 RA patients were treated with musculoskeletal ultrasound of 22 joints,including bilateral wrist,metacarpophalangeal and proximal interphalangeal.Then we evaluated the synovial hyperplasia of the above joints with gray-scale ultrasound,as well as analyzed the correlation between the joints score combinations of n different joints(denoted by GSUSn)and DAS28 and inflammatory cytokines.Results The levels of IL-6,TNF-?,IL-17 A,IL-1?and IL-1? in plasma of RA patients were significantly higher than those in healthy controls,and were positively correlated with DAS28?The data showed that the GSUS22,GSUS14(Bilateral wrist joint + MCPII-IV + PIPII-IV),GSUS12(Bilateral MCPII-IV + PIPII-IV),GSUS10(Bilateral wrist joint + MCPII-III + PIPIII-IV),GSUS8(Bilateral MCPII-III + PIPIII-IV),GSUS6(Bilateral wrist joint +MCPII-III)as well as GSUS4(Bilateral wrist joint +MCPII)were all positively correlated with DAS28.Moreover,GSUS14,GSUS10,GSUS8,GSUS6 and GSUS4 were positively correlated with plasma levels of IL-6,while there also existed positive correlations between GSUS12 and GSUS8 and IL-1? plasma levels.Conclusion GSUS4 not only reflects the disease activity of RA patients,but also indicate the plasma levels of IL-6.So we may conclude that bilateral wrist and MCPII should be the focus in the ultrasound examination.
