Objective To investigate the effect and mechanism of high mobility group protein B1-IL-1?complex on the expression of tumor necrosis factor-?and interleukin-6 in synovial fibroblasts of rheumatoid arthritis.Methods The synovial tissues of 10 patients with rheumatoid arthritis were isolated and cultured by digestive enzyme culture,and the fifth generation of rheumatoid synovial fibroblasts were used in the experiment.Stimulation of synovial fibroblasts from rheumatoid arthritis with different concentrations of HMGB1(0.1?1?10?100ng/m1)?IL-1?(0.01?0.1?1?10 ng/m1)?HMGBl/IL-1?complex(0.1/0.01?0.1/0.1?0.1/1?0.1/10?1/0.01?1/0.1?1/1?1/10?10/0.01?10/0.1?10/1?10/10?100/0.01?100/0.1?100/1?100/10 ng/m1).Detection of tumor Necrosis Factor-?and Interleukin-6 m RNA expression in synovial fibroblasts of Rheumatoid Arthritis by Real-time quantitative Polymerase chain reaction.Detection of TNF-?and IL-6 in the supernatant of synovial fibroblasts from patients with rheumatoid arthritis by enzyme-linked immunosorbent assay.Results Compared with the control group,the expression level of TNF-?m RNA and concentration of TNF-?in the IL-1?intervention group were higher than those in the control group(P<0.05);The expression of TNF-?m RNA and concentration of TNF-?in HMGB1/IL-1?complex intervention group was higher than that in HMGB1 intervention group(P<0.05);Compared with HMGB1 intervention group,the expression of TNF-?m RNA and concentration of TNF-?was increased in HMGB1/IL-1?complex intervention group with different concentration gradients(P<0.05);Compared with the IL-1?intervention group,the expression of TNF-?m RNA and concentration of TNF-?increased when IL1?increased to 1ng/ml and10ng/ml in each concentration gradient HMGB1/IL-1?complex intervention group,and the expression of TNF-?m RNA increased in a dose dependent manner with the increase of HMGB1 concentration(P<0.05);Compared with the control group,the expression level of IL-6 m RNA and concentration of IL-6 in the IL-1?intervention group were higher than those in the control group(P<0.05);The expression of IL-6m RNA and concentration of IL-6 in HMGB1/IL-1?complex intervention group was higher than that in HMGB1 intervention group(P<0.05);Compared with HMGB1 intervention group,the expression of IL-6 m RNA and concentration of IL-6 was increased in HMGB1/IL-1?complex intervention group with different concentration gradients(P<0.05);Compared with IL-1?intervention group,the expression of IL-6 m RNA and concentration of IL-6 was increased in HMGB1/IL-1?complex intervention group with different concentration gradients Conclusion 1.The formation of HMGB1-IL-1?complex can promote the secretion of TNF-?and IL-6,by RASFs,but the inflammatory ability of HMGB1 itself is very weak,and the induction of RASFs is weaker than that of IL-1?,indicating that HMGB1,as a co-inflammatory factor,cooperates with IL-1?to mediate the expression of TNF-?and IL-6 by RASFs.2.When the concentration of IL-1?in HMGB1-IL-1?complex increased to a certain range,the expression of TNF-?and IL-6 in RASFs increased in a dose dependent manner with the concentration of HMGB1.
