Objective:Glioma is the most common primary intracranial tumor,originates from glial cells.The highest incidence is glioblastoma?GBM,WHO??,accounting for46.1%,followed by diffuse astrocytomas.Treatment is based on surgical resection of tumors,combined with radiotherapy,chemotherapy and other comprehensive treatment.However,the prognosis of patients is generally poor.It is very important to find new molecular targets of predicting and prolonging the survival time of patients.Vitamin D can inhibit the proliferation and invasion of tumor cells,promote the apoptosis of tumor cells and inhibit the angiogenesis.Vitamin D need to combine with the vitamin D receptor?Vitamin D Receptor,VDR?to play its role.To investigate the roles of VDR in glioma,we detected the expression level of VDR protein and glioma associated protein ISO citrate dehydrogenase 1?IDH1?,tumor suppressor gene P53 and proliferation index Ki67 in glioma patients by immunohistochemistry,and analyzed the relationship between the expression of VDR and clinicopathological features and prognosis of glioma.Methods:Eighty cases of glioma were selected from the patients files of Shengjing hospital of China Medical University from January 1st,2013 to December 31st,2016.According to WHO classification,they were divided into 27 cases of low grade?WHO?-??,53 cases of high grade?WHO?-??.Immunohistochemical method was used to detect the expression of VDR and tumor associated protein IDH1,P53 and Ki67 in 80 cases of human glioma tissues by SP method,and the correlation analysis was made according to the clinicopathological characteristics and prognosis information.Results:The expression of VDR is higher in high grade glioma tissue than that in low grade glioma,the difference of expression levels of them are statistically significant.The expression of VDR is positively correlated with WHO grade?peri-tumoral edema?tumor necrosis,but not correlated with tumor size,location,age,sex,while the expression of VDR protein was negatively correlated with the expression of tumor associated protein IDH1,but was positively correlated with the expression of P53 and Ki67 protein.Kaplan-Meier survival curve analysis showed that prognosis of patients with high grade gliomas is poor with relatively low grade glioma patients?P<0.05?,the patients with the positive expression of IDH1 have better prognosis than the patients with negative expression of IDH1.The survival rate of patients with low expression of P53 and Ki67 is much higher than those of high expression,the survival rate of the patients with high expressions of VDR is lower than those with low expressions of VDR.Conclusion:Overexpression of VDR in high-grade glioma is correlated with tumor proliferation and development,it can be used as a poor prognostic marker.In addition,the VDR can also be used as a potential therapeutic target for the treatment of high-grade gliomas.
