Objective:To investigate the relationship between the expression of isocitrate dehydrogenase R132H(IDH1 R132H),hypoxia inducible factor-1?(HIF-1?)and vascular endothelial growth factor(VEGF)in gliomas and clinicopathological features.Methods:The location of gliomas was identified by brain CT scan,and the grade of gliomas was determined by hematoxylin-eosin(HE)staining,immunohistochemistry was used to detect the expression of IDH1 R132H,HIF-1?and VEGF in 51 glioma tissues and 10 normal brain tissues,and to analyze the relationship between them and the clinicopathological features of glioma.Results:According to the exclusion criteria,51 patients with glioma confirmed by surgery and pathology were selected.(1)Reading the CT of the brain to identify the locations of glioma tissues:14 cases of frontal lobe,13 cases of temporal lobe,5 cases of parietal lobe,6 cases of occipital lobe,13 cases with multiple sites(multisites were two or more intracranial sites).(2)After HE staining,HE was graded according to2016WHO classification and grading diagnostic criteria for central nervous system tumors:grade ? glioma 11 cases,grade ? glioma 20 cases,grade ? glioma 14 cases,grade ? glioma 6 cases.(3)The case group and the normal group were analyzed:the positive rate of IDH1 R132H,VEGF and HIF-1? in the case group was significantly higher than that in the normal group(P<0.05).Relationship between expression of IDH1 R132H,HIF-1?and VEGF protein and clinicopathological features:in 51 cases of glioma tissue,there were 21 positive expressions of IDH1 R132H,The positive expression rates of grades ?,?,? and ? were 27.27%(3/11),50.00%(10/20),57.14%(8/14),and 0.00%(0/6),respectively.Male and female positive expression.The rates were 50.00%(13/26)and 32.00%(8/25)respectively.The positive expression rates of<45 years old,45-59 years old and?60 years old were 35.71%(10/28),57.14%(8/14)and 33.33%(3/9),respectively.The positive expression rates in occipital lobe,parietal lobe,temporal lobe,frontal lobe and multiple sites were 50.00%(3/6),80.00%(4/5),46.15%(6/13),21.21%(3/14),38.46%(5/13),respectively.After statistical analysis,there was no difference in the expression of IDH1 R132H in the age,gender,grade and position groups.There were 24 positive expressions of HIF-1?,the positive expression rate was 47.06%(24/51),The positive expression rates of ?,?,? and ? were 45.45%(5/11),35.00%(7/20),50.00%(7/14)and 83.33%(5/6),respectively.The positive expression rates of male and female were 53.85%(14/26)and 40.00%(10/25),respectively.The positive expression rates of<45 years old,45-59 years old and?60 years old were 32.14%(9/28),57.14%(8/14)and 77.80%(7/9),respectively.The positive expression rates of occipital lobe,parietal lobe,temporal lobe and frontal lobe were 50.00%(3/6),40%(2/5),53.85%(7/13)and 21.42%(3/14),respectively.the positive expression of HIF-1? was different in middle age and old age in age group(P<0.05),regardless of gender,tissue grade and location.There were 31 cases with positive expression of VEGF,the positive expression rate was 60.78%(31/51),The positive expression rates of ?,?,? and ? were 72.73%(8/11),55.00%(11/20),64.29%(9/14)and 50.00%(3/6),respectively.The positive expression rates of male and female were 57.69%(51/26)and 64.00%(16/25),respectively.The positive expression rates of<45 years old,45-59 years old and?60 years old were 60.71%(17/28),57.14%(8/14)and 66.67%(6/9),respectively.The positive expression rates of occipital lobe,parietal lobe,temporal lobe and frontal lobe were 16.67%(1/6),40.00%(2/5),76.92%(10/13)and 50.00%(7/14),respectively.statistical analysis showed that VEGF was different in the location of glioma group(P<0.05);it was not related to gender,age and tissue grade.(4)Correlation analysis between the two indicators of IDH1 R132H,VEGF and HIF-1?:there was a statistically significant difference between VEGF and HIF-1? protein expression(?~2=3.89,P<0.05),but there was no statistically significant difference between IDH1 R132H and HIF-1?protein expression(?~2=0.004,P=0.948),and there was no statistically significant difference in VEGF protein expression(?~2=1.697,P=0.193).When the expression of IDH1R132H was negative,the positive expression of HIF-1?was 58.33%(14/24),and the positive expression of VEGF was 51.61%(16/31).Conclusion:1.HIF-1?expression in tumor tissues of glioma patients may be related to age,and VEGF expression may be related to glioma site.HIF-1?expression may be strongly correlated with VEGF expression in glioma.2.There are single or multiple positive expressions in the tumor tissues of glioma patients,and the combined detection of IDH1 R132H,HIF-1?and VEGF may provide clues for the diagnosis of glioma.
