| Objective: While the current methods of cancer treatment continue to develop,but the patients with cancer are still bothering of adverse reactions after treatment,such as surgery,postoperative adjuvant radiotherapy,combined with radiotherapy and chemoradiotherapy and targeted therapy,.Chemotherapy is used as a palliative treatment for patients with metastatic disease,and there is still a need to develop new and reasonable therapies for cancer.The new Phosphorus-magnesium whiskers have excellent mechanical properties and elastic moduli.They have been widely used in biomedical fields such as enhanced toughness,antibacterial,bone formation,and drug loading.However,research on cancer resistance is still insufficient.In this study,a series of in vitro experiments were performed on cells to evaluate the Impact on growth and apoptosis of tongue squamous cell carcinoma(Tca 8113)cells and mouse fibroblast(L-929)cells with different concentrations of new magnesium phosphite whisker suspensions,an exploratory study of the treatment of oral squamous cell carcinoma.Methods:1.Phosphorus magnesium whisker prepared into 1600 ppm,1400ppm,1200 ppm,1400ppm,1200 ppm,6 groups of different mass concentration of dilute suspension liquid,two kinds of cells on the orifice of inoculation and cultivation.2.CCK8 method is used to evaluate each whisker of Tca 8113 and L-929 cell proliferation,the effects of computing relative increment rate and cell toxicity analysis.3.Between groups on the cell morphology change under fluorescence microscope to observe cells.4.The Anexin V FITC/PI double dye test for two kinds of cell apoptosis.5.The influence of flow cytometry analysis PI single dye detection level of two kinds of cell cycle,to explore the influence of whisker content on the cycle distribution.And the results were analyzed.Results:1.CCK8 results showed that the increment rate decreases with the increase of concentration of whisker two cells,inhibition rate increased.When the concentration of whisker is less than 1200 ppm,non-toxic to L-929 cells;the non-toxic concentration is less than 1000 ppm to Tca 8113.So when the whisker concentration discussed is1200 ppm,tumor suppressor.2.On the cell morphology observation,Tca 8113 cells treated by different stimulus after 24 h,fluorescence microscope found that from 1200 ppm cell morphology,nucleus concentration and cell apoptosis.1400 ppm,1600ppm cell membrane rupture,likely necrosis.3.FCM to measure the cell cycle result,1200 groups of experimental whisker tca8113 cells concentration of cell cycle arrest in G1 phase,G1 phase cell proportion compared with the Empty vector group compared with obvious increase,and the S phase cell percentage decrease obviously.I showed the concentration of whisker make the head and neck squamous cancer cell cycle by G1 phase to S phase transformation process is restrained,the slow process of cell cycle.And L929 cell group as compared group,with 1200 ppm G1 phase and S phase almost no obvious change.4.The FCM measuring Annexin V FITC/PI double dye the experimental results show that when the whisker concentrations is 1200 ppm compared with negative control group,Tca8113 cellsearly apoptosis increased significantly,and L-929 almost had no effect.The Tca8113 early apoptosis cells number into bar chart,the percentage is as high as 69.6%.Conclusions:When whisker concentration is 1200 ppm can inhibit the growth of tumor cells,inducing apoptosis.It has potential significance for the prevention and cure of OSCC. |
