Objective: To study the effects and mechanism of the water,ethanol extract and different polar fractions of ethanol extract from Reineckia carnea on Antitussive,Expectorant,Antiasthmatic and Antipyretic,and analysis chemical components of volatile oil and petroleum ether,ethyl acetate 1-butanol and water polar fractions,for a further comprehensive understanding of Reineckia carnea and to provide experimental basic date.Methods: Extracts and polar fractions of Reineckia carnea were orally administrated to coughing mice induced by ammoniun hydroxide and coughing guinea pigs induced by capsaicin,using ELISA method for determination of substance P level change in serum and bronchoalveolar lavage fluid(BALF)of guinea pigs;injected with phenol red;sprayed to asthma guinea pigs induced by the mixed liquor with 2% acetylcholine chloride and 0.1% histamine diphosphate and OVA,using ELISA method for determination of IgE、INF-γ、NO、IL-4、IL-5、IL-13 level change in BALF of guinea pigs;administrated to fever rats by yeast and LPS,using ELISA method for determination of c AMP level change in serum of rats.Analysis the volatile chemical constituents of the whole plant,root and leaf of the dried and fresh products from Reineckia carnea were identified by GC-MS,and UPLC-Q/TOF-MS analysis was used to identity the petroleum ether,ethyl acetate 1-butanol and water polar fractions from the dried products.Results:1.The extract of Reineckia carnea has obvious inhibitory effect on coughing mice by chemical stimulation;it can increase the excretion of phenol red and increase the secretion function of respiratory tract in mice;it also can inhibit the asthma reactionof guinea pigs induced by the mixture of histamine phosphate and acetylcholine chloride;it can significantly reduce the fever and temperature of the rats induced by yeast and LPS,and inhibit the plasma c AMP level.It has good cough relieving,expectorant,antiasthmatic and antipyretic effects.From the experimental results,it was also found that the 75% ethanol extract of Reineckia carnea had better antitussive,expectorant and antiasthmatic efficacy than water extract.2.In the three polar fractions of PE,Et Ac,Bu OH,Et Ac and Bu OH can reduce the coughing times of capsaicin induced coughing in guinea pigs and prolong the latent period of coughing,which is the 75% ethanol extract of Reineckia carnea,it can significantly reduce the concentration of substance P in the serum and BALF,this suggesting that its antitussive effect may be related to the inhibition of the release of substance P by coughing C fiber receptor.Et Ac and Bu OH parts can increase the excretion of organ phenol red,reduce the viscosity of tracheal mucus and reduce phlegm.Bu OH parts can significantly prolong the incubation period of asthma in guinea pigs by the mixture of His and Ach,and inhibit the decrease of animal weight during the stage of asthma model stimulation.At the same time,the level of IgE,NO and IL-4 can be significantly reduced,the level of INF-γ can be significantly increased.It’s suggested that the location of Bu OH of the ethanol extract of Reineckia carnea may be inhibited by regulation the production and release of IgE,NO,IL-4,INF-γ,and inhibiting inflammatory factors to reach the antiasthmatic effect.3.By GC-MS analysis,55 kinds of volatile chemical constituents were identified from the dry Reineckia carnea,and 50 kinds from the fresh herb,the volatile components of the Reineckia carnea were mainly contain hydrocarbons,alcohols,ketones and oxygen containing heterocyclic compounds.By analysis of UPLC-Q/TOF-MS,143 compounds were identified from the ethanol extract of Reineckia carnea,containing a large number of carboxylic acid and carboxylic derivatives,saponins and amino acids.Conclusion: The extract of Reineckia carnea had significant antitussive,expectorant and antiasthmatic effect,ant had good antipyretic effect,and the effect of alcohol extract was better than water extract.The mechanism may be to relieve cough byreducing the release of substance P from C fibers,by increasing the excretion of phenol red and increasing the secretion function of respiratory tract,the viscosity of tracheal mucus decreased to achieve expectorant effect,by regulation the production and release of IgE,NO,IL-4 and INF-γ,the inflammatory factors can be suppressed to relieve asthma,by inhibiting plasma c AMP level and lowering body temperature,antipyretic effect can be achieved.
