Objective: Oral and maxillofacial malignancies have a high incidence and rank the 6th among system malignant tumors,of which oral cancer ranks the 12 th.Oral cancer has a high incidence in non-developed countries.It is estimated that there are about 500,000 new cases of maxillofacial and neck diseases in the world every year.According to reliable data,the incidence of oral cancer(oral and salivary gland)in the country is 0.19% for males and 0.16% for females.Oral squamous cell carcinoma is the most numberous in oral and maxillofacial cancer.Recent studies have shown that the incidence of oral squamous cell carcinoma was increased significantly these years.Relative to the world,in our country,a developing country,the incidence and mortality of oral squamous cell carcinoma is relatively high.The main reasons are the long duration,late discovery and poor prognosis.Salivary adenoid cystic carcinoma is painless in development of the tumor,featuring perinatal infiltration and multiple characters.Lymph nodes metastasis usually rarely occurred.Because of the metastasis occult,we cannot detectable neck metastasis.This tumor lacks detailed pathologic assessment of the lymph nodes due to the absence of neck dissection.However,hematogenous metastasis is common,especially lung,bone and liver.As the most common cancer of oral and maxillofacial surgery,we have made great progress in the treatment thearpy in recent years.However,the prognosis has not been greatly improved mainly due to their strong topo-infiltrability and high distant transfer rates.A better understanding of the molecular mechanisms associated with these tumors may be helpful in finding effective adjuvant therapy.Researchs about chemokines and their receptors mainly focused on inflammation.Now reports about them are mainly concentrated on the occurrence,recurrence and metatasis of various types of tumors.XCR1 is a kind of lymphoid chemokine receptors,a receptor for lymphotactin XCL1.There are not so many reports about XCR1 and oral cancer.Many reports confirmed that XCR1 is expressed in a variety of human cancers and correlated with the biological behavior of cancer cells.It needs further study whether XCR1 can improve the treatment effect of oral malignant tumors.In this study,we analyzed the expression of XCR1 in oral squamous cell carcinoma and salivary adenoid cystic carcinoma,as well as the relation with tumor differentiation,lymph node metastasis and TNM stage.In addition,we also used si RNA interference technology to transfect SACC-83 cells and performed a series of experiments on cell biology to explore the significance of XCR1 in salivary adenoid cystic carcinoma.The final results can clear the role and significance of XCR1 expression in SACC initially,which can indirectly provide a reference for XCR1 as a therapeutic target of SACC.1.Expression of XCR1 in oral squamous cell carcinoma and salivary adenoid cystic carcinoma and its relationship with the pathological parametersMethods: Paraffin-embedded specimens of 150 patients with oral squamous cell carcinoma undergone surgery in oral and maxillofacial surgery at Shengjing Hospital affiliated to China Medical University from 2009 to 2016 were selected,with adjacent tissues as OSCC control group.We also selected 40 paraffin block of salivary adenoid cystic carcinoma tissues,with parotid tumor resection of benign tumor paravertebral tissues as a control group.Immunohistochemistry was used to detecte the expression of XCR1 in oral squamous cell carcinoma and salivary adenoid cystic carcinoma.Results were statistically analyzed by SAS Medical Statistics software.Results: The expression of XCR1 in squamous cell carcinoma and parotid gland tissues was low.However,XCRi was highly expressed in cancer tissues.And there was a relationship between XCR1 expression and two kinds of cancer differentiation,as well as lymph node metastasis and TNM stage.Conclusion: 1,XCR1 is expressed in oral malignant tumors.2,There was a relationship between XCR1 expression and pathological parameters of oral malignancies.2.XCR1 expression in SACC-83 cells and biological behaviorMethods: RT-PCR method was used to verify the expression of XCR1 in SACC-83 cells,and the most effective si RNA was selected.CCK-8 was used to verify the difference of cell proliferation;cell scratch test was used to verify the change of cell migration rate;cell invasion assay was used to detect the invasion and migration of cells;Flow cytometry was used to confirm the change of apoptosis levels after XCR1 transfection.Results: PCR results showed that XCR1 knockdown efficiency.The results of CCK-8 showed that the cell proliferation after transfection decreased.The results of Transwell chambers and scratches showed that the invasion and migration of cells decreased after inhibiting the expression of XCR1.The rate of early and late apoptosis was up-regulated after XCR1 transfection.Conclusion: Proliferation,invasion and migration levels of SACC-83 cells with knocked-down XCR1 was decreased,while the rate of apoptosis increased.
