Salivary adenoid cystic carcinoma (SACC) is one of the common glandmalignancies in head and neck. The tumor is progressive invasion, typically withperineural extension. Distant metastasis to the lungs and bones hematogenic isfrequent. These features lead to a poor long-term survival rates after therapy. Becauseof the rudimentary understanding to the molecular mechanisms of SACC, the presenttherapy is now confine to the empirical surgery、radiation therapy and Chemotherapy,.The purpose of this study is to explore the complex gene regulation and change onmolecular level. By building upon this foundation, it is our hope that discoveries canbe contributed to the gene therapy.Our research group have found that the expression of Notch1and Notch4receptor is obvious different between two cell lines of SACC which are different inmetastasis[. In order to further understanding the relationship between the Notch1、Notch4and the metastasis and invasion of SACC, NICD1、NICD4recombinantadenoviral expressing vector was constructed. In vitro cell invasion assay and cellmigration assays were used to estimate the effect of NICD1and NICD4oninvasiveness of salivary adenoid cystic carcinoma and then the relative mechanismwas explored.The first part of this study was aimed to construct NICD1and NICD4recombinant adenoviral expressing vector. NICD1and NICD4gene was cloned intothe pShuttle-IRES-hrGFP-1vector and then homologous recombinanted withadenoviral backbone plasmid. Titered virions was acquired after transfection therecombinants into293A cell. SACC cells were infected with the virions, Western Blotwas used to confirm the expression of NICD1and NICD4protein in SACC cells.In the second part of this study, salivary adenoid cystic carcinoma cells whichwere infected with NICD1and NICD4recombinant adenovirus respectively wereused to cell proliferation assays, cell migration assay, cell invasion assay in vitro.In conclusion, the data indicate that the overexpression of Notch1and Notch4in salivary adenoid cystic carcinoma cells could promote the proliferation, migration andinvasion ability of SACC cells. Further investigations with Chip-seq will more closelyexamination the specific mechanism of the effects.
