Objective:To compare the exosomes secreted by adenoid cystic carcinoma cell lines and oral tongue squamous cell carcinoma.To investigate the effects of exosomes secreted by different cell lines of human salivary adenoid cystic carcinoma on proliferation,migration and invasion.Methods: ExoEasy Maxi Kit was used to extract exosomes.Using transmission electron microscopy to observe the morphology and diameter of exosomes.We also undergo the Nanoparticle tracking analysis(NTA)to identify the size and concentration of exosomes.Western Blotting was used to detect expression and semi-quantitative analysis of specific proteins in Sacc-83,Sacc-lm,Tscc-25,Tscca cell lines exosomes.CCK-8 method,plate colony formation assay,cell scratch and Transwell assay were used to detect ability of proliferation,migration and invasion.Results:Transmission electron microscopy and nanoparticle tracking analysis showed that exosomes are between 30-210 nm in diameter and the morphology and particle size were slightly different.Western Blotting detected the specific proteins of cell lines' s exosomes.The specific proteins CD9,CD63 and TSG101 were positively expressed and significant differences(P<0.01).NTA concentration analysis showed that the secretion quantity of exosomes about different cells.They were significantly different(P<0.01).The CCK-8 and plate cloning experiments showed that Sacc-derived exosomes enhanced the proliferation of cancer cells(P<0.05).Wound healing test and Transwell experiments showed that exosomes enhanced the migration and invasion of cancer cells(P<0.05).Conclusion: Both immortalized cell lines of two oral and maxillofacial malignant tumors can secrete exosomes,the morphology,particle size,specific marker proteins and secretion quantity are different.And the exosomes of salivary adenoid cystic carcinoma can enhance the proliferation,migration and invasion of cancer cells.
