Obejective:The TNBS/ ethanol induced UC model was used to observe the homing and inflammatory repair ability of MSCs from the peripheral blood of the UC model rats to the colonic mucosa tissue from the UC model of induced by the UC model.To compare the effect of Shen Ling Baizhu powder and Tong Xie Yao Fang on the homing of the peripheral blood MSCs to the colonic mucosal tissue and the repair ability of the inflammatory injury in the rat model of the model,the homing of the peripheral blood from the peripheral blood from the peripheral blood of the rat to the colonic mucosa and the repairing ability of the inflammatory injury.Objective to compare the intervention effects of Shen Ling Baizhu powder and Tong Xie Yao Fang on the peripheral blood MSCs homing and inflammatory injury repair ability of the model rats.Methods:1 Preparation of experimental drugs On the basis of Shenlingbaizhusan and Tongxieyaofang primary ratio,using the traditional Chinese medicine decocting method were concentrated to 2.26g/ml,0.78g/ml,placed in the refrigerator to spare.2 Establishment and verification of experimental animal model 88 rats of SPF grade healthy male SD were divided into blank group,model group,Shenlingbaizhusan group and Tongxieyaofang group with 22 rats in each group,72 rats were modeling,fasting 12 h,colon injection of TNBS ethanol solution to rats,then keep the natural sober,once a week,for 5 week.After the end of the model,2 blank groups and 2 model groups were randomly selected.They were executed quickly and cut off the colonic tissue.HE staining was used to evaluate the success of the model.3 Colonization of BMSCs in rats The colonization of the colonic mucosa in the rat was observed by DAPI labeled BMSCs by laser confocal microscope.The pathological changes of colon tissue in each group were observed by HE staining,and the influence of the two sides on the homing of BMSCs from peripheral blood to colonic mucosa and the repair ability of inflammatory injury in the UC rats were compared.4 Two parties promote the homing of BMSC to UC colon mucosa Westernblotting to detect the expression of SDF-1/CXCR4 protein in the colonic mucosa of each group,and RT-PCR to detect the expression of SDF-1/CXCR4 m RNA in the colonic mucosa of each group.Result: 1 After the injection of BMSCs through tail vein,the colonic mucosa of each group was repaired as follows: naked eye observation: compared with the blank group,the model group had severe erosion,congestion and edema,and thickened and rigid bowel wall.There are multiple surfaces of ulcers.The injury score increased,with significant difference and statistically significant(P<0.01).Shenlingbaizhu group compared with the model group,colonic mucosal hyperemia,the adhesion degree was reduced slightly.The number of ulcers decreased and the injury score decreased,and the difference was statistically significant(P<0.05).Tongxieyaofang group and model group compared with colonic mucosal edema,congestion,erosion degree is reduced considerably,injury score was decreased,the difference was statistically significant(P<0.01);compared with Shenlingbaizhusan group,colonic mucosal adhesion decreased,the difference was statistically significant injury score(P<0.05).2 Confocal laser display results: compared with the blank group,the model group increased the content of BMSCs,with the difference between the two groups(P<0.05),Shenlingbaizhusan group and Tongxieyaofang group content of BMSCs was increased,with the difference(P<0.01);compared with the UC model group,Slingbaizhu group and pain laxative groups BMSCs levels were increased.,with significant difference(P<0.01);compared with Shenlingbaizhu group,Tongxieyaofang group increased the content of BMSCs,with a significant difference(P<0.01).3 3 the results of the detection of SDF-1/CXCR4 protein in the colonic mucosa of each group showed that,compared with the normal group,the SDF-1 of the colon mucosa tissue of the model rats was significantly increased(P<0.05),and the expression of CXCR4 protein was not significantly increased(P> 0.05).The expression level of SDF-1 and CXCR4 protein was significantly higher in the group of Shen Ling Baizhu San and Tong Xie Yao Fang(P<0.05),and it was better than the group of Shen Ling Baizhu powder.The results of SDF-1/CXCR4 m RNA of colon mucosa tissue of rats in each group were detected by 4 RT-PCR method.Compared with the normal group,the expression of SDF-1 and CXCR4 m RNA in the colon mucosa tissues of the model rats were obviously up-up,and the difference was statistically significant(P<0.05).Compared with the model group,the expression level of SDF-1 and CXCR4 m RNA in the colon mucosa tissue of the group of Shen Ling Baizhu powder group and the Tong Xie Yao Fang Group was obviously up,and the pain Xie Yao Fang was obviously superior to the group of Shen Ling Baizhu powder(P<0.05).Conclusion:UC inflammatory injury can promote the homing of the peripheral blood BMSCs to the colonic mucosa,but it does not participate in the repair of the injury of the mucosa of the colon of UC,and both the Shen Ling Baizhu powder and the Tong Xie Yao Fang can improve the damage of the colonic mucosa tissue of the UC rats.The mechanism may be associated with the increase of the expression of SDF-1 and CXCR4 in the colon mucosa and the promotion of the peripheral blood BMSCs to the colon.The homing of the mucous membrane.In the above intervention effect,the effect of the Tong Xie Yao Fang Group is obviously superior to the group of Shen Ling Baizhu powder.According to the theory of "corresponding prescription of TCM syndrome",the model of UC rats induced by TNBS/ ethanol method may be related to the syndrome of liver depression and spleen.
