| Ulcerative colitis is a chronic non-specific ulcerative colitis,its etiology agentsit has not been found clear,mucosal and submucosal inflammation is the main characteristics of this disease,ulcerative colitis and crohn's disease referred for inflammatory bowel disease.Because this is difficult to cure,high ratio of multicentric,the dysplasia is relatively high,therefore the world health organization called it is one of modern disease that is difficult to cure.The current medication includes salicylic acid,corticosteroids,immunosuppression and surgical treatment,etc,although various treatments can relieve symptoms, there are some minus effect and complications,curative effect is not complete, illness is recurrence easely.How to improve inflammatory bowel disease treatment,reduce adverse reactions,is an important problem in clinical urgently to be solved.The traditional Chinese medicine thinks that it occurs due to inadequate innate endowment,including improper diet,The traditional Chinese medicine think that there are many factors of disease cause,weakness of the spleen and the stomach,external contraction of the six excesses,dietary irregularities,affect-mind dissatisfaction,and constitutional insufficiency can cause ulcerative colitis by harming the function of spleen-stomach,these can cause spleen,stomach,kidney and other organs dysfunction,qi and blood stasis,bowel mucosa changed the symptom such as purulent blood into damaged.Traditional Chinese medicine has a long history and rich experience, practice has proved that Traditional Chinese medicine prevention with definite effect less adverse reactions,safety and enduring,these worth we summarize and research.This subject will use cell and molecular biology technology and theory to compare and healing efficacy of TCM therapy about induction of mesenchymal stem cells(MSCs) in repairing UC,that would raise the clinical curative effect of traditional Chinese medicine to ulcerative colitis.Objective:To explore the effect of induction of Mesenchymal stem cells transplantation in rats UC intestinal positioning and different TCM therapy about induction of mesenchymal stem cells(MSCs) in repairing UC,this paper discusses intervention,comparison and selection of the TCM therapy effectively.Methods:According to the principle of randomized,female rats were divided into six groups,the normal group,the model group,the MSCs transplantion group,the Shenlingbaizhu decoction group,the Taohongsiwu decoction group,the Jinkuishenqi decoction group,every group has 20 rats. Another one male SD group has 20 rats to cultivate MSCs.In addition to the normal group,the remaining rats in each group before the UC model made can not eating but water lasting 48h,let the rats struggle and defecate the distal colon excrement 2 times,each time interval 6h above.With chlorine ammonia ketone,intraperitoneal injection of anesthetized rats with silicon tube inserted in rats,the anus 8cm about TNBs 10s,tube into finished.After the separation of cultivating the male rat bone marrow stromal cells,by end of intravenous transplanted it into the female rats,give different traditional Chinese herbs lavage observe different effect of traditional Chinese medicine intervention. Gastric lavage animal doses,according to experimental animals and measurement of body surface area ratio conversion,the Shenlingbaizhu decoction group doses to 30g/kg/only,the Taohongsiwu decoction group doses to 12g/kg/only,the Jinkuishenqi decoction group doses to 16g/kg/only, once-daily treatment for 2 weeks.By immunocytochemistry appraisal MSCs phenotype,using PCR detection colon tissue specific gene male sry segment. Using immunohistochemistry to test in rats transplantion MSCs group and the rats transplantion MSCs group treated with herbal decoction colonic mucosa intestinal tissue Stem cells markers such as Musashi-1 and TERT.Results:(1)By immunocyto chemistry,separation and in vitro culture MSCs appraisal phenotype,CD44 phenotype expressed is positive,CD34 phenotype expressed is negative.Compared with the normal group,each group intestinal mucosa's damaging index increased significantly,showed that the experimental ulcerative colitis animal models are maded successful.(2)After traditional Chinese medicine intervention,MSCs transplantation group and herbal treatment group were reduced mucosa damage index,Taohongsiwu decoction group is significantly reduced mucosa damage index(P＜0.05).(3)After using PCR detection,detected colon tissue male gender gene segments sry,transplantion MSCs group positive rate is 20%,Shenlingbaizhu decoction group positive rate is 25%,Jinguishenqi decoction group positive rate is 25%,Taohongsiwu decoction group positive rate is 35%.(4)Using immunohistochemistry to test in rats transplantion MSCs group and the rats transplantion MSCs group treated with herbal decoction colonic mucosa intestinal tissue Stem cells markers such as Musashi-1 and TERT,PCNA increased obviously,Taohongsiwu decoction group is significantly increased(P＜0.05).Traditional method to promote blood is more effective in MSCs proliferation.Conclusion:MSCs transplantation in receptor UC rats have positioning in intestinal,traditional Chinese medicine has certain intervention Stem cell proliferation especially the therapeutic method about promoting blood circulation and removing blood stasis.Taohongsiwu decoction stands for the therapeutic method about promoting blood circulation and removing blood stasis,this method can repair,and accelerate UC colonic mucosa ulcer healing. this kind TCM therapy is effective.
