Objective The skin ?-degree deep burn scalding model was developed in Kunming mice.To investigate the therapeutic effect and the mechanism of lavender oil on ?-degree deep burn wound healing and scar prevention..Methods32 Kunming mice,were separated as 6 in the blank,6 in model,10 in control and 10 in test group.After mice were anesthetized,depilated with 8% sodium sulfide solution on upper back.Thermostatical water bath with 95 ?,put mice hair removal area on the steam port upper 10 cm for 10 s.Scald ?-degree deep burn scar model was established.The model,control and test group were treated with saline,hirudoid cream and lavender oil respectively,twice a day for 28 days.The wound healing and scar formation were recorded.In each group,mice were taken blood from their tails on 7th day and eyeballs 28 th day;the serum was removed after centrifugation.Scar tissue was biopsied on 7th and 28 th day,ELISA was performedto detect the serum level of tgf-?1 ? Type I Pro-collagen ? Type III Pro-collagen and IL-10.On the 28 th day of treatment,scar tissue were cut down and histological exmination was made.The histological changes in wound healing and scar formation were observed.The thickness of epidermis,thickness of scar and hair follicle were recorded.SPSS 16.0 was performed for data analysis,data was expressed by mean ±standard deviation,and t-test and one-way anova were for comparison between groups.When the homogeneity of variance was homogeneity,variance analysis was used to analyze the mean of multiple groups.When the variance is different,Kruskal-Wallis was used.Therefore,the LSD method was to compare the skin and scar thickness.The Kruskal-Wallis test was to compare the hair follicle indicators,index description was described by M(p25-p75).The differences were statistically significant with P < 0.01.Results1.The result of ?-degree deep burn model:1.1 The clinical skin wound and histopathological manifestations of ?-degree deep were observed model group.The wounds process includes inflammatory reaction,granulation tissue hyperplasia and scab;the scar formed finally.1.2 The TGF-beta 1 and IL-10 in serum,the ratio of type I and type III pro-collagen in tissues of blank group were 64.3±9.25ng/ml?48.48±3.70ng/ml?1.35±0.04.2.The outcome of treatment:2.1 The model group was purple and red in color,irregularin shape,touch hard and rough in surface.The degree of inflammation was heavy and healing slowlly.The epidermis,scar and hair follicle were sharply hyperplasia.The thickness was78.3±36.0?m in epidermis,1129.2±495?m in scar;the number of hair follicles in the unit area was 3.5.TGF-beta 1,IL-10,the ratio of type I /III pro-collagen on 7th day after modeling were 129.19±10.29ng/ml?58.54±4.17ng/ml?4.02±0.24;On 28 th day were 249.80±7.35ng/ml?67.25±2.83ng/ml?7.31±0.47,and the values was gradually increased,and significantly higher than the normal level(P<0.01).2.2 The control group was pink in color,more regular in shape,touch soft and smooth in surface.The degree of inflammation was lighter and healing quickly.The epidermis,scar and hair follicle were less hyperplasia.The thickness was 74.4±20.6?m in epidermis,970.6±301.7?m.1 in scar,the number of hair follicles in the unit area was3.5.TGF-beta 1,IL-10,1,the ratio of type I/ III pro-collagen on 7th day after modeling were 265.58±9.95ng/ml?73.64±8.10ng/ml?3.64±0.33;On 28 th day were116.40±19.78ng/ml ? 59.70±3.59ng/ml ? 2.29±0.29,and the values was increased significantly on 7th day,and slightly higher than the normal level on 28 th day(P<0.01).2.3 The test group was bright red in color,most regular in shape,touch soft and smooth in surface.The degree of inflammation was light and healing quickly.The epidermis,scar and hair follicle were weakly hyperplasia.The thickness was 54.3±13.?m in epidermis,424.5±119.5?m in scar,the number of hair follicles in the unit area was 1.TGF-beta 1,IL-10,1,the ratio of type I /III pro-collagen on 7th day after modeling were 303.57±8.03ng/ml?88.08±3.90ng/ml?3.20±0.27;On 28 th day were69.45±11.31ng/ml ? 50.06±6.16ng/ml ? 1.79±0.51,and the values was increased dramatically on 7th day,and declined to normal level on 28 th day(P<0.01).Conclusion1.We developed a ?-degree deep burn model via Kunming mice,and which can simulate the characteristic changes of human skin scald features.2.Lavender oil significantly promote wound healing and inhibit scar in mice with ?-degree deep burn.The level of TGF-beta 1,IL-10,and the ratio of type I/ III pro-collagen were regulated significantly.
