Proliferative viteroretinopathy(PVR) and proliferative diabetic retinopathy (PDR)were two kinds of fiber proliferative diseases that were usually seen in eyes.Theirclinical feathures were recurrent and bad prognosis. The vision of the patients wasdeclined seriously or even blind.PVR and PDR had the similar pathologicalcharacteristics.There were fiber proliferative membrances on the surface of the retinaand fiber cords below the retina in PVR and PDR patients.The retina was tracted bythe fiber proliferative membrances and fiber cords result in retina detachment.PVRand PDR were usually cured difficultly in clinic.There were differences between PVRand PDR,the new vessels could be found in the roliferative membrances of PDRpatients,but could not be found in PVR. Transforming growth factor-β2(TGF-β2) andtype I collagen(COL-1) were deemed to take part in the forming of extracellularmatrix(ECM) and new vessels.So it is benefit for exploring the mechanism of PVRand PDR to detect TGF-β2and COL-1in proliferative membrances on the surface ofretina.PurposeTo explore the effect and mechanism of TGF-β2and COL-1in PVR and PDR by detecting the expression of TGF-β2and COL-1and their relationship in proliferativemembrances of PVR and PDR patients.MethodsHematoxylin-eosin(HE) staining and immunohistochemistry(IHC) was used tostain and detect the the expression of TGF-β2and COL-1in proliferativemembrances of PVR and PDR patients from vitrectomy,including control group6eyes,PVR/C group13eyes,PVR/D group11eyes,PDR group12eyes.ResultsThe results of HE staining were as follows:both kinds of the proliferativemembrances were composed of Retinal pigment epithelial(RPE) cells、Fibroblast andCollagen fibers. The results of IHC staining were as follows: the TGF-β2and COL-1were rarely seen in the control group, the TGF-β2was expressed in the cytoplasm andthe surface of cell membrance,the COL-1was expressed in the ECM of theproliferative membrances. The average optical density of TGF-β2and COL-1were asfollows(X±S): control group was54.62±8.37,63.32±7.81,PVR/C group was178.25±11.30,198.74±16.23,PVR/D group was229.17±14.25,287.33±9.78,PVRgroup(PVR/C group and PVR/D group) was203.71±12.24,243.04±13.05,PDRgroup was203.84±7.82,247.81±11.17. Statistical analyses were performed by usingSPSS17.0software. The average optical density of TGF-β2and COL-1in PVR/Cgroup、PVR/D group and PDR group were significantly higher than control group(P<0.05). The average optical density of TGF-β2and COL-1in PVR/D group weresignificantly higher than PVR/C group (P<0.05).There were not statistical differenceof the average optical density of TGF-β2and COL-1between PVR group and PDRgroup(P>0.05).The expression of TGF-β2and COL-1in the proliferativemembrances of PVR and PDR patients was strongly correlative(r=0.775,P<0.05). Conclusion1.TGF-β2and COL-1took part in the formation of the proliferative membraneof PVR、PDR together.2.TGF-β2and COL-1presented a trend of increasing with the progress of PVR,the expression of TGF-β2and COL-1in the proliferative membrances of PVR andPDR patients was strongly correlative.
