Objective:To investigate the correlation studies between the changes of lymphocyte subsets and IL-17 levels in peripheral blood of patients with advanced non-small cell lung cancer after chemotherapy.Methods:From October 2016 to February 2018,65 patients with advanced non-small cell lung cancer who were admitted to the Medical Oncology Department of Second Affiliated DaYi Hospital of Shanxi Medical University were selected as the study subjects(NSCLC group).In the same period,30 healthy persons from the hospital were selected as the control group.Flow cytometry was used to compare the distribution of peripheral blood total T cells(CD3+),helper T cells(CD3+CD4+),inhibitory/cytotoxic T cells(CD3+CD8+),and CD4+/CD8+ ratio,cytotoxic T cells(CD8+CD28+,CTL,TC),suppressor T cells(CD8+CD28-),regulatory T cells(CD4+CD25HiCD127Lo,Treg)and natural killer cells(CD3-CD56+,NK),the measured by ELISA to detect the distribution of IL-17.At the same time,gender,age,different clinical stages(? B,IV),different pathological stages,and before and after treatment(after chemotherapy 2 cycles)the number of lymphocyte subsets in peripheral blood of patients with advanced non-small cell lung cancer was analyzed,and a correlation analysis between CTL?CD8+CD28-cells and IL-17 was performed.Results:(1)Compared with the control group,the serum level of IL-17 in the advanced NSCLC group was significantly higher,and the level of IL-17 in the peripheral blood of the NSCLC group was significantly lower than that before the treatment(P<0.05).(2)Compared with the control group,the ratio of CD3+ cells,CD3+CD4+ cells,CD4+/CD8+,CD8+CD28+ cells,and NK cells in the peripheral blood was significantly decreased in the NSCLC group,while the proportion of CD3+CD8+ cells,CD8+CD28 cells,and Treg cells was significantly increased.The difference was statistically significant(P<0.05).After treatment,the proportion of CD3+ cells,CD3+CD4+ cells,CD4+/CD8+,CD8+CD28+ cells,and NK cells increased significantly in the NSCLC group after treatment,whereas the proportion of CD8+CD28-cells and Treg cells increased.Significantly decreased,the difference was statistically significant(P<0.05).(3)Pearson correlation analysis suggested that IL-17 levels in patients with advanced non-small cell lung cancer were negatively correlated with the ratio of CTL(r=-0.650,P<0.05),and there was no correlation between IL-17 level and CD8+CD28-ratio(r=-0.034,P>0.05).Conclusion:(1)The levels of IL-17 in peripheral blood of advanced NSCLC were higher than those in normal subjects.There was a difference in IL-17 levels before and after treatment,suggesting that it may serve as an indicator for lung cancer monitoring.(2)The dynamic monitoring of lymphocyte subsets in patients with peripheral blood may provide clinical guidance for the analysis of disease status and prognosis in patients with advanced NSCLC.(3)The level of IL-17 in peripheral blood of patients with lung cancer was negatively correlated with CTL.
