[Objectives]To investigate the distribution of innate-like lymphocytes in peripheral bloods of normal persons and non-small cell lung cancer (NSCLC) patients.[Methods]Six fluorescence staining and flow cytometry were used to detect the percentages of iNKT lymphocytes, γδT lymphocytes, B1 lymphocytes and adaptive T lymphocyte, B2 lymphocytes in peripheral blood lymphocytes of 16 normal persons (Group C),5 NSCLC fist-visit patients (Group F) and 15 stable state NSCLC patients after treat (Group S).[Results]1. The percentages of iNKT lymphocyte in peripheral blood lymphocyte of Group C, Group F and Group S were (0.31±0.42)%, (0.17±0.12)%, (0.06±0.07)%, respectively; the percentages of γδT lymphocyte were (5.09±4.08)%, (6.26±4.22)%, (2.96±2.37)%, respectively; the percentages of B1 lymphocyte were (1.97±0.88)%, (1.04±0.74)%, (1.50±1.48)%, respectively; the percentages of adaptive T lymphocyte were (57.98±11.98)%, (65.68±10.64)%, (71.64±11.17)%, respectively; the percentages of B2 lymphocyte were (7.76±3.20)%, (10.02±6.66)%, (7.52±4.85)%, respectively.2. Spearman correlation analysis was used to analyze the correlation relationship between the percentages of several lymphocyte subsets. There were positive correlation relationships between iNKT cell and y8T cell in Group S (r=0.5336, p=0.0426); B1 cells and B2 cells in Group C and Group S, respectively (r=0.7964,p=0.0006; r=0.6029,p=0.0153).3. We also found that the percentage of iNKT cell of Group F was significantly higher than Group S (p=0.026); the percentage of γδT cell of Group F was significantly higher than Group S (p=0.040); the percentage of iNKT cell of Group S was significantly lower than Group C (p=0.031), though the percentage of adaptive T lymphocyte was much higher (p=0.003); the percentage of B1 cell of Group F was significantly lower than Group 0=0.046).[Conclusions]1. The percentages of innate-like lymphocyte in all the samples were far below the adaptive immune cells, with great rang of change, individual difference was obviously. Dynamic monitoring the percentages of innate-like lymphocyte subsets was of active advantages in treatment and development of tumors.2. The connection of innate immune and adaptive immune may be precisely reflect on the quantitative correlation between the subsets of innate-like lymphocyte.3. The B1 cells and B2 cells of both controls and stable patients were significantly positive correlation, but fist-visit patients were not, maybe prompted that B subsets of the fist-visit patients were imbalance.4. Compared with stable patients, fist-visit patients had significantly higher percentages of both iNKT cell and γδT cell. This phenomenon suggested that patients after treatment with increased percentages of iNKT cell and γδT cell might be risk factors for the development of malignant tumor, rather than consequences following tumor.5. Compared with controls and stable patients, the latter had a significantly higher percentage of adaptive T cells. It was possible that the tumor infiltrating adaptive T cells backflowed to peripheral blood caused the rise up of percentage, or the high variational number of total white blood cells of tumor patients themselves covered the truth of change of cell subsets. This suggested that when analyzed lymphocyte subsets of tumor patients, it is important to pay attention to the absolute number of lymphocytes in addition to the percentages of cells.
