Study On The Quality Standard And Analgesic Activity Of Resina Draconis

Resina Draconis,the name of deep red resin extract from Dracaena cochinchinensis(Lour.)S.C.Chen.which belongs to the family of Liliaceae has been applied in clinical medicine treatment of hurts,inflammation,stomach ulcers,diarrhea and diabetes.This paper took Resina Draconis as research objective,researched on chemical constituents and analgesic pharmacological activities of total flavonoids extract.On the basis of analysing chemical constituents about Resina Draconis,the chemical fingerprint analysis method was investigated and quality standard was estabished.First of all,the chemical constituents of Resina Draconis had been isolated systematically.23 compounds,including 15 flavonoids,5 stilbenes,1 lignans,1 chlorine-containing compound and 1 simple phenolic compound,was identified as 7,4?-dihydroxy-8-methylflavan(1),Kumatakenin B(2),Pinoresinol(3),Resveratrol(4),Loureirin D(5),7,4?-dihydroxy-5-methoxydihydrohomoisoflavonoid(6),7,4?-dihydroxydihydrohomoisoflavone(7),Loureirin C(8),4,4?-dihydroxy-2,6-dimethoxydihydrochalcone(9),7,4?-dihydroxyflavan(10),1,2-dihydroxy-4-methoxybenzene(11),5,7,4?-trihydroxy-8-methylflavan(12),Pinostilbene(13),3,5-dihydroxystilbene(14),Cochinchinenin B(15),7-hydroxy-4'-methoxy-8-methylflavan(16),Loureirin A(17),Loureirin B(18),4?-hydroxy-3?,7-dimethoxyflavan(19),Pterostilbene(20),3,5-dimethoxystilbene(21),2,3,5,6-Tetrachloro-1,4-dimethoxyben zene(22),4?-Hydroxy-7-methoxy-8-methylflavan(23).Secondly,the weakness of the general identification items,inspection items and content determination items had been perfected compared with original national standards.9 batches of Resina Draconis from 3 different places were collected as samples.To establish TLC identification methods,loureirin A,loureirin B and cochinchinenin B were used as control with samples.The establishment of UltravioletVisible Spectrophotometry Determination of total flavonoids,the content of total flavonoids of sample were measured in 39.98 %~49.87 % with loureirin B as control.According to the four-part of Chinese Pharmacopoeia in 2015 Edition,the contents of water,ash and alcohol insolubles were measured in 3.01 %~5.49 %,0.33 %~0.76 % and 1.51 %~1.94 %,respectively.And the content of heavy metal was less than fifteen per million.Through high performance liquid chromatography,the contents of loureirin A,loureirin B and cochinchinenin B were measured in 0.14 %~0.99 %,0.54 %~1.99 % and 0.12 %~0.84 %.In this paper,a fingerprint evaluation method which objectively reflects the internal quality of Resina Draconis was established and provides a basis for quality evaluation and quality standard improvement.What's more,the mice's acetic acid twisting model and hot plate model were used to determine the analgesic activities of total flavonoids extract.The results indicated that the extract had marked inhibitory effects on pain caused by chemical stimulation(acetic acid)and heat stimulation(hot plate).In the writhing analgesic experiments,it showed strongest analgesic effect when given 75 mg/kg dosage.In the mouse hot plate analgesia experiment,37.5 ~ 150 mg/kg extract has obvious analgesic activity,compared with the positive drug.
