Objective.Prolactin-secreting adenomas(prolactinomas)is a kind of benign tumor accounted for approximately 40% of all pituitary tumors.According to the statistics,the incidence of prolactinomas varies with sex and age,females between 20 and 40-years-old in significantly higher than other groups.Because of hyperprolactinemia,the characteristic clinical manifestations of these patients is menstrual disturbances(usually amenorrhea)and galactorrhea.The pathogenesis of prolactin-secreting pituitary adenomas is complicated and not clear so far.HPGD is a gene that located on chromosome 4 in humans and encodes a 29 kD enzyme named 15-hydroxyprostaglandin dehydrogenase(15-PGDH).This enzyme involved in prostaglandin catabolism and catalyzes the NAD?-dependent oxidation of the 15(S)-hydroxyl group of lipoxins and prostaglandins resulting in the production of 15-keto-lipoxins and 15-keto-prostaglandins,which exhibit greatly reduced biological activities.15-hydroxyprostaglandin dehydrogenase is widely distributed in various mammalian tissues,but the role in pathological consequences including cancer remains undefined.It is investigable to determine if the expression of 15-hydroxyprostaglandin dehydrogenase is altered during prolactin-secreting pituitary adenomas tumorigenesis.In the present study,we demonstrate that the expression of HPGD mRNA and 15-hydroxyprostaglandin dehydrogenase in clinical specimens and normal pituitaries in order to evaluate the effects of HPGD on prolactin-secreting pituitary adenomas tumorigenesisMethods.In the present study,we demonstrate the expression of HPGD mRNA and 15-hydroxyprostaglandin dehydrogenase in clinical specimens and normal pituitaries through qRT-PCR and Western blot analysis.Results.The quantitative reverse transcription polymerase chain reaction analysis of HPGD expression in 20 pituitary adenomas tissues and 3 normal pituitary tissues indicated that the expression of HPGD mRNA levels in normal tissues was each higher than that in tumor tissues.It was significant difference in HPGD mRNA levels between the tumor group and the control group(P<0.05).The Western blot analysis of 15-hydroxyprostaglandin dehydrogenase expression in 20 pituitary adenomas tissues and 3 normal pituitary tissues indicated that the expression of 15-hydroxyprostaglandin dehydrogenase levels in normal tissues was each higher than that in tumor tissuesConclusions.In summary,We confirm that HPGD behaves as a tumor suppressor through encode 15-hydroxyprostaglandin dehydrogenase in prolactin-secreting pituitary adenomas.
