Objective: To compare and analyze the perfusion characteristics and differences between CXCL12 antibody targeting contrast agent and conventional contrast agent(sonovine)in ovarian cancer transplantation tumor at different stages,and to further explore the application value of targeted ultrasound contrast agent.Methods: Fifty mice bearing ovarian cancer xenografts were randomly divided into 5 groups,10 days,20 days,30 days,40 days and 50 days after transplantation,one group of them were selected to study the characteristics of targeted ultrasound contrast(MBt)and general ultrasound contrast(MBc),blood perfusion images and time signalparameters were obtained,including peak intensity(PI),peak time(TTP)and mean transit time(MTT),Statistical differences of MBt and MBc parameters within the same group and between different groups were analyzed.According to the characteristics of contrast-enhanced ultrasound perfusion and the results of contrast-enhanced imaging parameters,the targeted ultrasound groups at 40 and 50 days were divided into the non-necrotic tumor group(n=8)and the partial necrotic tumor group(n=12)according to the standard of whether the tumor had necrosis(no blood perfusion area).Results: The values of PI and MTT of targeted contrast in the 5 groups were higher than those of normal contrast group,and the TTP values of targeted contrast agent in the 20 day and 30-day groups were lower than those of normal contrast agent,with statistically significant difference(P<0.05).Comparison of the contrast parameters of targeted contrast agents between different groups showed statistically significant differences(P<0.05),Pairwise comparison showed the highest PI value in the 30 d group,and the 30 day group had statistically significant differences from the 10 day,40day and 50 day groups(P<0.05).There was no statistically significantdifference between PI and MTT values in the MBc group(P>0.05).The values of PI and MTT in the non-necrotic tumor group were higher than those in the partial necrotic tumor group,and the difference was statistically significant(P<0.05).Conclusions: Targeted ultrasound contrast agent with CXCL12 is superior to conventional contrast agent,providing a basis for the early diagnosis and targeted treatment of ovarian cancer.Advanced tumor necrosis may be one of the important factors affecting tumor perfusion.
