Objective:To investigate the applicability of targeted ultrasound contrast agent microbubbles with vascular endothelial growth factor receptor type2in angiogenesis in ovarian transplantation tumor of nude mice. Methods:The transplant tumor model of SKOV3in nude mice was established and MBt was prepared by biotin-avidin conjugation method;Immunofluorescence ment Immunofluorescence method are used to observe the combination of ultrasound microbubble surface of monoclonal antibodies; Mice with tumor were treated with MBt and control microbubbles for ultrasound contrast examination and relative parameter analysis of time-intensity curves (TIC); VEGFR2expressions in angiogenesis of tumor tissues were analyzed by immunohistochemistry. Results:Nude mouse carrying ovarian cancer SKOV3cell transplantation tumor model was established successfully; Immunofluorescence observed under fluorescence microscope microbubble ultrasound surface bright green fluorescence; MBt carrying with VEGFR2was prepared successfully; MBt group73.95(69.50,83.08) dB has higher peak intensity (P<0.01) than MBc group57.55(54.30.74.10) dB, MBt group480.00(442.50,617.50) S has longer lasting time (P<0.01) than MBc group354.00(297.50,412.50) S; Immunohistochemistry detected high VEGFR2expression in angiogenesis endothelium of tumor tissues. Conclusions:MBt with VEGFR2can bind with angiogenesis endothelium in ovarian transplantation tumor of nude mice and achieve targeted ultrasound molecular imaging. MBt achieves better performance in early diagnosis of ovarian cancer than MBc.
