Objective:Neurodegcnerativc diseases are one of the major diseases that endanger the health of middle-aged and elderly people,among which Alzheimer's disease(AD)and Parkinson's disease(PD)are the most common.At present,most of the drugs used in clinical treatment of neurodegenerative diseases are chemical drugs.Nerve cell transplantation and kidney-tonifying traditional Chinese medicine in the treatment of neurological diseases have become the focus of research in the treatment of neurological diseases in recent years due to their long-lasting effects and small side effects.In cell transplantation therapy,neural progenitor cells(NPCs)is commonly used to transplant seed cells,is a kind of ability to self-renewal and multiple differentiation potential of the nerve cells,but from embryonic or nerve tissue NPCs in the clinical application of cell source is also less,organize immune rejection,ethics and other issues,so by in vitro induced to obtain high purity and high active NPCs is the key to solve this problem.In recent years,traditional Chinese medicine for tonifying the kidney has also achieved remarkable results in the treatment of neurological diseases.Therefore,the screening of Chinese medicine with neuroprotection also provides theoretical basis for the treatment of patients with neurological diseases.Methods:In this paper,mice induced pluripotent stem cells(miPSCs)were first induced to differentiate into NPCs cells in vitro,during which Retinoic Acid(RA),Epidermal growth factor(EGF)and Basic fibroblast growth factor(bFGF)were added,differentiation potential of the induced NPCs cells was identified;NPCs cells were used to screen the activity of traditional Chinese medicine extracts for tonifying kidney under 2D conditions;NPCs cells were cultured in 3D using polyvinyl alcoho/sodium alginate/velvet antler hydrogel complex.The biocompatibility of the complex hydrogel to NPCs cells was observed by microscope and staining.In addition,CORT disease model was established under 3D conditions to detect the protective effect of velvet antler polypeptide on NPCs cells under 3D conditions.Results:The results showed that the NPCs cells induced in vitro showed positive expression of PAX6 and could be further differentiated into oligodendrocyte progenitor cells(OPCs).Chinese medicine extracts for tonifying the kidney under 2D conditions.In the experiment of NPCs cell proliferation with Chinese medicine extracts under 2D conditions,different Chinese medicine extracts can promote cell proliferation to varying degrees within the concentration range.In the experiment of NPCs cell disease injury protection with Chinese medicine extracts,cortisone(100?M)and glutamate(1 mM)were used to damage NPCs cells,and two in vitro models of simulated neural disease were established.The cell survival rate was determined by MTT assay.The results showed that pilose antler extract had a protective effect on CORT injury of NPCs cells in the range of 0.25-1 mg/ml concentration,while epimedium herb had a protective effect on CORT injury of NPCs cells in the range of 50-100?g/ml concentration.In the Glu injury model,velvet antler extract had a protective effect on NPCs cells at 0.5 mg/ml.The results of 3D culture showed that the hydrogel complex had good biocompatibility with NPCs,and it could protect and promote the differentiation of NPCs cells into oligodendrocyte precursor cells(OPCs),neurons and astrocytes under the disease model.Conclusion:In summary,miPSCs can be successfully induced into NPCs,and the induced NPCs can be used to screen the activity of Chinese herbal extracts.Polyvinyl alcohol/sodium alginate/velvet antler polypeptide hydrogel complex has good biocompatibility with NPCs cells under 3D culture conditions,can protect and promote the proliferation and differentiation of NPCs. |
