Morehella esculenta,one kind of rare edible and medicinal fungus,belongs to phylum of Ascomycota,class of Pezizomycetes,order of Pezizales,family of Morchellaceae,genus of Morchella.It is popular with a large amount of nutritional substance,such as polysaccharide,protein,and trace elements.M.esculenta polysaccharides(MPS)from have lots of bioactivies,including anti-hyperlipidemia,antioxidant and antitumor etc.However,little research concerning enzyme-assisted M.escuenta polysaccharide(EnMPS)from the fruiting bodies has been conducted so far.In this work,the extraction conditions of MPS were optimized using Plackett-Burman(PB)test and response surface methodology(RSM)test.Meanwhile,the monosaccharides composition,molar ratio,infrared spectroscopy antioxidative,anti-hyperlipidemia and organ protection of EnMPS were investigated as well as MPS was also analyzed.The present conclusions were listed as follows:The temperature,extracting time,solvent/material ratio,multiple,ethanol concentration,pH value and ethanol time were selected by PB test from M.esculenta by water extraction and alcohol precipitation.The results showed that the optimum conditions of MPS extraction were ethanol time(12.06h),solvent/material ratio(1:30.02)and extracting time(3.00h)by RSM,the maximum theory rate of MPS extraction reached 1.30%.Under the condition of ethanol extraction time(12.00h),solvent/material ratio(1:30.00)and extracting time(3.00h),the actual rate was 1.26%.The results of gas chromatograph showed that EnMPS contained five different monosaccharides including rhamnose,xylose,mannose,glucose and galactose with a mass percentage of 4.90%,16.82%,16.40%,54.42% and 74.5% and with the molar ratio of 1:3.76:3.05:10.13:1.39,while the MPS was consisted of four different monosaccharides including rhamnose,xylose,mannose and galactose with a percentage of 2.19%,33.89%,17.37% and 46.55% and with the molar ratio of 1:16.92:7.23:19.36.The infrared spectral experiments showed that both EnMPS and MPS were furan sugars.In this work,the effects of EnMPS or MPS on superoxide,hydroxyl and 1,1-diphenyl-2-pyrazole hydrazide(DPPH)radicals were analyzed.The scavenging rates of EnMPS at 600mg/L on superoxide,hydroxyl and DPPH radicals reached 76.92±2.61%,66.74±2.56% and 75.78±2.4%,respectively.It had been confirmed that EnMPS showed superior antioxidant effects than MPS in vitro.Animals experiments suggested that the EnMPS exhibited superior abilities of reducing hepatic lipid levels by monitoring the serum enzyme activities(ALP,ALT and AST)and serum lipid levels(CK,TC,TG,HDL-C,LDL-C and LDH),enhancing the kidney index(UREA,CREA and ALB),the hepatic antioxidant enzymes(FFA,SOD,CAT and T-AOC)and decreasing the lipid peroxidation(MDA and MPO).The histopathological sections also gradually became normal with the intervention of polysaccharide,indicating that EnMPS and MPS had protective effects on the organ injuries.The above results demonstrated that EnMPS had protect organic effects by antihyperlipidemia and antioxidation in High-fat-high-cholesterol emulsion induced mice,which might be exploited as a potential natural and active health food for organic protection and its complications.
