Objective:Using active ingredients,serum pharmacochemistry and pharmacokinetic methods,the pharmacodynamic material basis of Weichangshu tablets was studied.Methods:1.Screening effective active sites of Weichangshu tablets,silica gel column chromatography,gel permeation chromatography and high performance liquid chromatography were used to separate monomer components from effective parts.The components and struc tures of the compounds were identified by nuclear magnetic resonance.Then we detect the effects of these compounds on gastrointestinal motility in rats with gastric motility disorder.2.The serum samples of rats in the blank group and the Weichangshu tablets group were taken,HRMS was used to detect and identify the blood transitional components after oral administration of Weichangshu tablets in rats.3.HPLC-MS/MS was used to detect plasma samples of rats in each group,DAS 2.0 was used to calculate the pharmacokinetic parameters of two groups of rats.Two groups of parameters were compared by SPSS.Results:1.The effective parts of Weichangshu tablets were chloroform extraction locations,ethyl acetate extraction locations and 20%ethanol elution parts.14 compounds were isolated from the effective part of Weichangshu tablets,these are chrysophanol(?),rheochrysidin(?),emodin(?),formononetin(?),aloe emodin(?),rhein(?),benzoyl paeoniflorin(?),isoglycyrrhizin(?),glycyrrhizin(?),liquiritin(?),paeoniflorin(?),hydroxy paeoniflorin(?),albiflorin(??),glycyrrhizic acid(??).All compounds promoted gastric emptying rate and intestinal propulsion rate in rats with gastric motility disorders.2.The chromatographic conditions of the fingerprint in vitro were used,31 blood transitional components were identified,27 from the compound itself,and the other 4 compounds as the metabolites of the compound.3.The pharmacokinetic parameters of the glycyrrhizin,isoglycyrrhizin,formononetin,emodin,rhein and aloe emodin in the plasma of normal rats and gastrointestinal motility disorder rats were significantly different(p<0.05).Conclusion:To clarify the effective parts of the Weichangshu tablets,the material basis of Weichangshu tablets in vivo was basically elucidated.It laid a foundation for further study on the mechanism of action of Weichangshu tablets.
