Dongguan Hospital of Traditional Chinese Medicine Aijun Zhou,chief physician,has been using Xiao Chaihu Decoction as basic prescription,adding Dried ginger,Coptis chinensis,Poria cocos,Fried atractylodes to compose Modified Xiao Chaihu Decoction.And it is a classic prescription of Shanghan Lun written by Chinese medical sage Zhongjing Zhang.In recent 10 years,using Modified Xiao Chaihu Decoction to treat hundreds of gastric ulcer and chronic gastritis patients with remarkable curative effect.In order to provide the modern scientific basis for the clinical curative effect of modified Xiao Chaihu Decoction,our laboratory in cooperation with the Dongguan Hospital of Traditional Chinese Medicine applying to the Dongguan Municipal Science and Technology Bureau for the "modified Xiao Chaihu Decoction on pharmacodynamic material basis" research project.This thesis studies the effective parts of antibacterial and anti-inflammatory from modified Xiao Chaihu Decoction.Researching the componential analysis of physiological disposition on main effective chemical parts by serum pharmacochemistry to explain the material basis of drug efficacy.Under the guidance of the theory of traditional Chinese Medicine,considering the Modified Xiao Chaihu Decoction as a whole.Extracting and separating it to volatile oils,alkaloids,phenolic acids,saponins,polysaccharides.Studying the pharmacological tests of anti Helicobacter pylori on different chemical substances with Agar diffusion and liquid dilution methods.The result showed that chemical parts such as volatile oils,alkaloids,phenolic acids,saponins have different degrees anti-bacterial effects.By xylene induced mouse auricle swelling model and carrageenan vegetable gum induced rat paw edema model,to the site of different chemical substances for the pharmacological screening of and the results showed that the phenolic acids,saponins and polysaccharides have anti-inflammatory effects.Therefore,this study provides a scientific experimental basis for the clinical curative effect of Modified Xiao Chaihu Decoction.Investigating the physiological disposition of antibacterial and antiinflammatory chemical parts of Modified Xiao Chaihu Decoction with Serum Pharmacochemistry Method.By using liquid chromatography-mass spectrometry(LC-MS)analysis method for identification of the serum containing phenolic acid sites,separating and identifying eight constituents such as Oroxylin-A-7-O-glucuronic acid,trihydroxy-monomethoxyflavone glucuronic acid and wogonoside migrating to blood(see chart 6).The serum containing saponins sites were analyzed,separating and identifying twelve constituents such as Chrysin-6-C-?-D-pyran glucose-8-C-?-L-pyran arabinose,Oroxylin-A-7-O-glucuronic acid,baicalein-5-O-glucuronic acid,glycyrrhizin and glucuronic acid conjugates,baicalein and baicalein and glucuronic acid conjugates(see chart 8).The results proved that effective chemical parts of Modified Xiao Chaihu Decoction produced effect after getting into the blood.The degree thesis uses the interdisciplinary research method to explain the material basis of drug efficacy of Modified Xiao Chaihu Decoction,thus provide the modern scientific basis for the clinical curative effect,and lay a certain foundation for the development of new traditional Chinese medicine.
