Objective:To investigate the mechanism of the adenosine in the treatment of migraine,injecting the adenosine A1 receptor agonist(R-phenylisopropyl-adenosine)into the lateral ventricles of rats with nitroglycerin induced migraine.Methods:Choose male SD rats,about 200g,rates were taken into 6 groups,for example blank control group,model group,R-PIA0.5 group,R-PIA1.0 group,R-PIA2.0 group,R-PIA2.0+DPCPX(adenosine A1 receptor antagonist)group.Observed the numbers of the rats scratching their heads,climbing cage times and heart rate changes,and the ELISA,immunohistochemistry,Western blot techniques were used to detect the expression of the calcitonin gene-related peptide(CGRP)in the places of the blood,the trigeminal ganglion(TG),spinal trigeminal nucleus caudalis(TNC).Results:(1)Injection of R-PIA 0.5-2.Oug into the lateral ventricles can reduce the numbers of the rats cratching their heads and the times of the rats climbing cages,which had an analgesic effect on migraine model raand had no effect on the heart rate of the rat and the analgesic effect can be blocked ts,in a dose dependent manner,by DPCPX;(2)Injection of R-PIA into the lateral ventricles can reduce the expression of the CGRP in the blood,TG,TNC parts.Conclusion:Adenosine A1 receptor agonist can inhibit the activation of trigeminal vascular system by inhibiting the neurogenic inflammation reaction and trigeminovascular nociceptive transmission,which can produce analgesic effect on migraine;...
